细小病毒反向遗传操作技术研究进展
Advance in Reverse Genetics of Parvovirus
【摘要】 细小病毒感染的动物宿主广泛,包括猫科、犬科、猪科、牛科、鸭科和鼬科等动物,是动物的一种重要病原。病毒反向遗传学技术是在获得病毒全基因组序列的基础上,构建感染性克隆,通过DNA重组、定点突变、插入、缺失等遗传修饰手段创造突变体,并研究突变体所造成的表型效应,从而研究基因的生物学功能。论文对ssDNA细小病毒反向遗传学技术的研究进行综述,为细小病毒同科病毒反向遗传学操作平台的建立提供参考。
【Abstract】 Parvovirus is an important pathogen of animals which can infect a wide range of animals including Felidae,Canidae,Suidae,Bovidae,Anatidae,Mustelidae and other animals.Viral reverse genetics is constructing viral infectious clones by viral genomic sequence obtained firstly.Then viral infectious clones are used to study the biological function of genes and the phenotypic effect caused by mutants,such as DNA recombination,site-specific mutation,insertion,deletion and other genetic modification.This review mainly focussed on the advanced research in the field of single strand DNA parvovirus reverse genetics in order to provide a reference for the establishment of reverse genetics platform of homologous parvovirus.
