转录组的黄山栾树MYB基因家族鉴定及表达

【摘要】 植物叶片颜色是有色素组成、含量及分布动态决定,MYB转录因子在这一生物过程中发挥重要作用。为了揭示黄山栾树黄叶突变体‘金焰彩栾’叶片呈色的转录调控机制,依据转录组数据,通过挖掘MYB转录因子和生物信息学分析,结合不同组织表达对其功能进行分析。结果表明:从黄山栾树转录组共鉴定97个MYB转录因子,预测编码蛋白质所含氨基酸数目52~1 780个,分子质量大小52.0~193.35 ku。序列比对及进化分析发现,黄山栾树MYB基因与拟南芥MYB基因聚为23个亚家族,预测分别参与次生代谢、生长发育和胁迫响应等过程。结合表达特征分析,预测KbMYB80、KbMYB91、KbMYB6、KbMYB8和KbMYB40可能参与黄酮和花青素的生物合成调控,KbMYB60负调控花青素生物合成,KbMYB16和KbMYB88参与黄山栾树叶片细胞次生壁合成过程。

【Abstract】 The composition, content and distribution of pigments determined the color of plant leaves, and MYB transcription factor plays an important role in this biological process. In order to reveal the transcriptional regulation mechanism of Koelreuteria bipinnata ‘jinyan’, with the transcriptome sequencing(RNA-Seq) data, the MYB genes of K. bipinnata were identified. The results showed that 97 KbMYB transcription factors were excavated, and they encoded 52-1 780 amino acids, the molecular weight was 52.0-193.35 kDa. By phylogenetic analysis, MYB genes of K. bipinnata and Arabidopsis thaliana clustered into 23 subfamilies, which were predicted to be involved in secondary metabolism, growth and development, stress response and other processes. Additionally, differential expression data based on fluorescence quantitative PCR predicted that KbMYB80, KbMYB91, KbMYB6, KbMYB8 and KbMYB40 might be involved in the regulation of the biosynthesis of flavonoids and anthocyanins, KbMYB60 negatively regulated the biosynthesis of anthocyanins, and KbMYB16 and KbMYB88 were involved in the secondary wall synthesis process of the leaves of K. bipinnata.
