棕榈酸对绵羊单核巨噬细胞炎性因子和氧化应激水平的影响

【摘要】 棕榈酸(Palmitic acid,PA)是一种典型长链饱和脂肪酸,具有一定脂毒性,可引起细胞内过度炎症反应和氧化应激。研究旨在揭示PA对绵羊单核巨噬细胞中炎性因子表达和氧化应激水平影响,探讨TLR4基因介导作用,分析亚麻酸(Linoleic acid,LNA)可能存在的缓解效应。结果表明,在1 mmol·L-1PA处理4 h后,TLR4基因的表达水平显著上调(P<0.05);IFN-γ、IL-1β、TNF-α、IL-6等炎性因子表达水平显著高于对照组(P<0.05);ROS和iNOS等氧化相关指标显著升高(P<0.05),SOD、CAT、T-AOC等抗氧化相关指标显著降低(P<0.05)。利用TAK242抑制TLR4基因后,其表达水平显著下调(P<0.05),TNF-α和IL-6炎性因子表达水平极显著下调(P<0.01),ROS和iNOS表达水平下降、T-AOC表达水平升高(P<0.05)。PA与LNA共处理后,TLR4基因表达水平显著下调(P<0.05),TNF-α和IL-6炎性因子表达水平极显著下调(P<0.01),ROS和iNOS表达水平下降、T-AOC表达水平升高(P<0.05)。研究表明,PA处理后绵羊单核巨噬细胞炎性因子和氧化应激水平升高,TLR4基因在此过程中起重要介导作用,LNA可能在一定程度上缓解PA诱发的细胞炎症反应和氧化应激。

【Abstract】 Palmitic acid(PA), as a typical long-chain saturated fatty acid, has a strong lipotoxic effect and can cause excessive inflammatory response and oxidative stress in cells. This study aimed to reveal the effects of PA on the expression of inflammatory cytokines and oxidative stress levels in sheep monocytes macrophages, exploring the role of TLR4 gene, and analyzing the possible mitigating effect of Linoleic acid(LNA). The results showed that after 4 h with 1 mmol · L-1 PA treatment, the expression level of TLR4 gene was significantly up-regulated(P<0.05), the expression levels of inflammatory cytokines IFN-γ, IL-1β, TNF-α, and IL-6 were significantly higher than those in the control group(P<0.05). The levels ROS and iNOS were significantly increased(P<0.05), and the levels of antioxidant index SOD, CAT, and T-AOC were significantly decreased(P<0.05). After TAK242 treat-ment, the expression level of TLR4, TNF-α, and IL-6 were significantly down-regulated(P<0.05), the levels of ROS and iNOS were significantly decreased(P<0.05), and the level of T-AOC was significantly increased(P<0.05). After PA and LNA co-treatment, the expression level of TLR4, TNF-α, and IL-6 were also significantly down-regulated(P<0.05), ROS and iNOS levels were significantly decreased(P<0.05), and the T-AOC level was significantly increased(P<0.05). Our results demonstrated that the levels of inflammatory cytokines and oxidative stress in sheep monocytes macrophages were increased after PA treatment, TLR4 gene played a mediating role in this process, and LNA may alleviate PAinduced inflammatory response and oxidative stress.
