【摘要】 目的检测LncRNA TCONS₀0022381在胃癌组织中的表达,探讨其对胃癌SGC-7901细胞增殖的影响。方法 qPCR法检测LncRNA TCONS₀0022381（TCONS）在胃癌和癌旁组织中的表达;将胃癌细胞系SGC-7901细胞分为siRNA干扰组、阴性对照组和空白对照组,采用脂质体2000转染法将靶向TCONS的siRNA和阴性对照si RNA分别转染,空白对照组不予任何处理。转染48h后采用q PCR法进行抑制效率的鉴定;MTT法检测各组细胞增殖情况;Western blot法检测各组细胞增殖核抗原PCNA的表达情况。结果 LncRNA TCONS₀0022381在胃癌组织中的相对表达显著高于癌旁组织,差异有统计学意义（P<0.05）。LncRNA TCONS₀0022381的si RNA能够显著抑制其在SGC-7901细胞中的表达（P<0.05）。MTT实验可见siRNA干扰组较阴性和空白组相比,增殖活力显著下调（P<0.05）,PCNA的表达显著下调（P<0.05）。结论 LncRNA TCONS₀0022381在胃癌组织中显著高表达,对胃癌细胞具有促进增殖的作用。更多还原

【Abstract】 Objective To detect the expression of LncRNA TCONS₀0022381 in gastric cancer and its effect on proliferation of SGC-7901 cells. Methods The expression of LncRNA TCONS₀0022381 was determined by real-time quantitative PCR in gastric cancer samples and cell lines. The gastric cancer SGC-7901 cells were grouped into interference group, negative control group and blank group. The gastric cancer cells transfected with siRNA targeted to LncRNA TCONS₀0022381 was designated as siRNA interference group, while named those transfected with scrambled siRNA as negative control group and the cells without any treatment as blank group. After transfection of 48 hours, real-time quantitative PCR was used to detect the suppression of LncRNA TCONS₀0022381. The proliferation of each group was detected by MTT assay. The expression of proliferating nuclear antigen（PCNA） in each group was detected by Western blot. Results LncRNA TCONS₀0022381 was up-regulated in both gastric cancer tissue samples and cell lines（P<0.05）. si RNA transfection could significantly reduce the expression of LncRNA TCONS₀0022381 in SGC-7901 cells（P<0.05）. Compared with the negative and blank groups, the MTT assay showed that the proliferation activity and the PCNA expression were significantly down-regulated（P<0.05）. Conclusion LncRNA TCONS₀0022381 is highly expressed in gastric cancer and play a role in promoting the proliferation of gastric cancer cells.更多还原
