基于16S rDNA技术研究黄连解毒汤对db/db糖尿病小鼠肠道菌群的影响

【摘要】 目的:研究黄连解毒汤对db/db小鼠肠道菌群的影响。方法:将db/m小鼠作为空白组,db/db小鼠随机分为模型组、黄连解毒汤组和二甲双胍组;给药12周后,全自动生化仪检测血浆中血糖血脂指标[血糖(GLU)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)],使用16S rDNA技术检测各组小鼠粪便肠道菌群变化及预测db/db小鼠肠道菌群相关的功能通路。结果:与模型组比较,黄连解毒汤组体质量、TG和LDL-C水平降低(P<0.05);黄连解毒汤组肠道菌群从门到属水平均发生了变化,有益菌的丰度增加,有害菌的丰度降低,其中,Parabacteroides_goldsteinii、多形拟杆菌、纤线杆菌目、革兰菌属和鼠乳杆菌显著增多;共发现35条功能通路与db/db小鼠肠道菌群相关,其中碳水化合物代谢、能量代谢和糖合成与代谢等通路与糖尿病相关。结论:黄连解毒汤具有改善db/db糖尿病小鼠肠道菌群失调的作用。

【Abstract】 Objective: To investigate the effects of Huanglian Jiedu Decoction on gut microbiota modulation in db/db mice. Methods: The db/m mice were used as the control group and db/db mice were randomly divided into model group, Huanglian Jiedu Decoction group and Metformin group. After intragastric administration for twelve weeks, the blood glucose and lipid index were analyzed by an automatic biochemical analyzer, the changes of intestinal microflora were determined and functional pathways related to intestinal flora in db/db mice were predicted by 16S rDNA sequencing technology. Results: Compared with the model group, Huanglian Jiedu Decoction could effectively reduce the level of weight, triglyceride(TG) and low density lipoprotein-cholesterol(LDL-C)(P<0.05); the intestinal flora of Huanglian Jiedu Decoction group had changed from phylum units to the genus units, with increasing the abundance of beneficial bacteria and reducing the abundance of harmful bacteria, among them, the abundance of Parabacteroides_goldsteinii, bacteroides thetaiotaomicron, ktedonobacterales, gramella and lactobacillus murinus were significantly increased. A total of 35 functional pathways were found to be related to the intestinal flora of db/db mice, of which carbohydrate metabolism, energy metabolism, glycan biosynthesis and metabolism pathways were related to diabetes. Conclusion: Huanglian Jiedu Decoction has the effect of improving the intestinal microflora imbalance in db/db mice.
