桂枝汤调节LIF改善心脏交感神经胆碱能性质转化的作用机制
Study on the mechanism of Guizhi Decoction in regulating LIF to improve cholinergic transdifferentiation of cardiac sympathetic nerves
【摘要】 目的:探讨桂枝汤抑制心力衰竭心脏交感神经胆碱能性质转化的可能作用机制。方法:36只Dahl盐敏感(DS)大鼠随机分为模型组和桂枝汤不同桂芍比例组(桂芍比分别为1∶1、2∶1、1∶2,即桂枝汤1、2、3组),另取9只SS-13BN大鼠设为空白对照组。心脏超声检测大鼠心脏左心室射血分数(LVEF)、左室短轴缩短率(LVFS)指标变化,RT-PCR检测大鼠左心室白血病抑制因子(LIF)mRNA的表达,Western Blot和双抗体免疫荧光分别检测大鼠左心室酪氨酸羟化酶(TH)、胆碱乙酰转移酶(CHAT)蛋白及荧光表达。结果:与空白对照组比较,模型组LVEF、LVFS水平降低,LIF mRNA表达上升,TH蛋白和TH+阳性神经纤维表达下降,TH+/CHAT+阳性神经纤维水平下调(P<0.01)。与模型组比较,桂枝汤1组、2组LVEF、LVFS水平增加,LIF mRNA表达下降,TH蛋白和TH+阳性神经纤维表达上升,TH+/CHAT+阳性神经纤维水平上调(P<0.01,P<0.05),其中桂枝汤1组作用最佳。结论:桂枝汤桂芍1∶1、2∶1配伍可抑制心脏交感神经胆碱能性质转化,提高交感神经功能学支配,改善心功能,其机制可能与调节心脏LIF的表达有关。
【Abstract】 Objective: To explore the possible mechanism of Guizhi Decoction inhibiting the cholinergic transdifferentiation of cardiac sympathetic nerves in heart failure. Methods: Thirty-six salt-sensitive DS rats were randomly divided into model group and Guizhi Decoction group(with different proportions between Ramulus Cinnamomi and Radix paeoniae Alba at 1∶1, 2∶1, 1∶2, namely Guizhi Decoction 1, 2, 3 groups),9 SS-13 BN rats were selected as control group. Cardiac echocardiography was used to detect the changes of cardiac function of left ventricular ejection fraction(LVEF) and left ventricular short axis shortening rate(LVFS). The changes of leukemia inhibitory factor(LIF) mRNA were detected by RT-PCR. Western Blot and double antibody immunofluorescence were used to detect the expression of tyrosine hydroxylase(TH), choline acetyltransferase(CHAT) protein and fluorescence in left ventricle of rats in each group. Results: Compared with the control group, the levels of LVEF and LVFS decreased, LIF mRNA expression increased, TH protein and TH+ positive nerve fibers decreased, and TH+/CHAT+ positive nerve fibers decreased in model group(P<0.01). Compared with the model group, the levels of LVEF and LVFS level increased, LIF expression decreased, TH protein and TH+ positive nerve fibers increased, and TH+/CHAT+ positive nerve fibers increased in Guizhi Decoction 1, 2 groups(P<0.01, P<0.05). Moreover, Guizhi Decoction 1 group was better than Guizhi Decoction 2 group. Conclusion: Guizhi decoction with the proportions between Ranulus Cinnamomi and Radix paeoniae Alba at 1∶1 and 2∶1 could inhibit cholinergic transdifferentiation of cardiac sympathetic nerves, improve the innervation of sympathetic nerve function and cardiac function. The mechanism may be related to the regulation of LIF expression in the heart.
