解毒通络方对心肌成纤维细胞Vimentin和α-SMA表达影响

【摘要】 目的:探讨解毒通络方对大鼠心肌成纤维细胞增殖和转分化的影响。方法:以MTT法检测各组培养细胞增殖活力,碱裂解法测定各组培养液羟脯氨酸含量;RT-q PCR法检测培养细胞及Ⅰ型胶原(ColⅠ)mRNA表达;Western Blot和RT-qPCR检测Vimentin、α-SMA蛋白及mRNA表达。结果:模型组心肌成纤维细胞增殖活力、羟脯氨酸含量及ColⅠmRNA表达量均显著高于空白对照组,经解毒通络方及卡托普利处理后则显著降低(P<0.01)。模型组心肌成纤维细胞Vimentin、α-SMA蛋白及mRNA表达水平显著上调,解毒通络方及卡托普利处理后则显著降低(P<0.05,P<0.01)。结论:解毒通络方可抑制心肌成纤维细胞增殖活力及胶原等细胞外基质(ECM)合成和分泌,可能与其抑制Vimentin和α-SMA的转录和翻译有关。

【Abstract】 Objective: To discuss the effects of Jiedu Tongluo Formula on the rat cardiac fibroblasts. Methods: MTT method was used to analyze the proliferation activity of cells in each group, hydroxyproline content in cell culture medium was determined by alkaline hydrolysis method, Vimentin and α-smooth muscle actin(α-SMA) expression were detected by both Western Blot and real-time reverse transcript PCR(RT-qPCR), and the collagenⅠ(ColⅠ) mRNA was also detected by RT-qPCR. Results: The proliferation activity, the concentration of hydroxyproline and the level of ColⅠ mRNA were all increased in the model group significantly(P<0.01), but decreased statistically in both the various dose of Jiedu Tongluotreated groups and the captopril-treated group compared with that in the model group(P<0.01). In the model group, the protein and mRNA level of Vimentin, α-SMA were significantly up-regulated, and in Jiedu Tongluo-treated groups and the captopril-treated group they were decreased statistically(P<0.05, P<0.01). Conclusion: Jiedu Tongluo Formula can inhibit the proliferation and the ECM expression of the myocardial fibrosis which may have a relation with inhibition of Vimentin and α-SMA expression.
