D-甘露糖对炎性条件下牙龈间充质干细胞迁移、增殖和凋亡的影响

【摘要】 目的评估在体外D-甘露糖对牙龈卟啉单胞菌(Pg)超声提取物提供的炎性条件作用下的牙龈间充质干细胞的迁移、增殖和凋亡能力的影响。方法体外培养人牙龈间充质干细胞,用D-甘露糖预处理细胞72 h,在Pg超声提取物作用36 h后观察,用划痕实验检测细胞的迁移能力变化;用CCK-8法检测细胞增殖活性;用流式细胞术检测细胞凋亡。结果在Pg超声提取物提供的炎性条件下,D-甘露糖促进人牙龈间充质干细胞迁移;不影响牙龈间充质干细胞增殖;抑制了牙龈间充质干细胞凋亡。结论 Pg超声提取物可抑制牙龈间充质干细胞的迁移并诱导其凋亡,D-甘露糖可减弱这种作用。

【Abstract】 Objective To evaluate the effect of D-mannose on the migration, proliferation and apoptosis of gingival mesenchymal stem cells under the inflammatory conditions. Methods Human gingival mesenchymal stem cells were cultured in vitro. The cells were pretreated with D-mannose for 72 hours, and then observed with ultrasonic extract of Porphyromonas gingivalis for 36 h. The migration ability of the cells was detected by scratch test. CCK-8 assay was used to detect cell proliferation activity. Cell apoptosis was detected by flow cytometry. Results Mannose pretreatment promoted the migration of human gingival mesenchymal stem cells under the intervention of ultrasonic extract of Porphyromonas gingiva. Mannose pretreatment did not affect the proliferation of human gingival mesenchymal stem cells under the action of ultrasonic extract of Porphyromonas gingiva. Mannose pretreatment inhibited the apoptosis of human gingival mesenchymal stem cells induced by the ultrasonic extract of Porphyromonas gingiva. Conclusions The ultrasonic extract of Porphyromonas gingiva can inhibit the migration of human gingival epithelial cells and induce their apoptosis. D-mannose attenuates this effect.
