【摘要】 为了验证Cldn4(Claudin 4)蛋白在胰腺癌中扮演的角色是促进肿瘤的进展还是抑制肿瘤的进展,选用胰腺癌细胞PANC-1,分别对其Cldn4进行过表达和敲低,研究其对胰腺癌细胞迁移、增殖的影响.实验方法采用体外划痕实验及Transwell小室实验评估细胞的迁移能力,采用MTT比色法评估细胞增殖能力.结果表明:Cldn4可显著抑制PANC-1的迁移,但对其增殖能力无显著性影响,Cldn4在胰腺癌中扮演癌基因的角色.更多还原

【Abstract】 To test whether the protein Cldn4(Claudin 4) plays a role in promoting or inhibiting tumor progression in pancreatic cancer, pancreatic cancer cells PANC-1 were selected as the cell model for phenotype investigation. Overexpression and knockdown of Cldn4 stable cell line were established for tumor malignancy phenotype, cell migration and proliferation, and evaluation in pancreatic cancer. Wound healing and transwell assay were selected for evaluation of tumor metastasis ability. MTT statistics indicated the cell proliferation capacity. Experiment results show that Cldn4 inhibits the migration of PANC-1 but has no significant effect on proliferation of PANC-1. Cldn4 plays an oncogene role in pancreatic cancer.更多还原
