琴叶榕中佛手柑内酯的定性鉴别与含量测定

【摘要】 目的建立琴叶榕中佛手柑内酯的定性鉴别和含量测定方法,并对琴叶榕和全缘榕根、茎、叶部位的佛手柑内酯含量进行比较分析。方法采用TLC进行定性鉴别,并采用HPLC进行含量测定。色谱柱为Agilent ZORBAX Eclipse Plus C18(250 mm×4.6 mm,5μm),流动相为甲醇-水(55∶45),流速为1 m L/min,检测波长为246 nm,柱温为30℃,进样量10μL。结果琴叶榕的TLC图斑点清晰、分离度好;佛手柑内酯检测浓度在2.12~63.6μg/m L范围内与峰面积呈良好的线性关系(r=0.999 9),精密度、稳定性、重复性试验的RSD<2%;平均回收率为99.8%,RSD值为1.12%(n=6)。结论所建立的方法简便易行、准确、重复性好,可用于琴叶榕的质量控制。

【Abstract】 Objective To establish the method for qualitative identification and content determination of bergapten, and to comparative analysis the content of the different parts for roots,stems and leaves in Ficus pandurata Hance. Methods TLC was adopted for qualitative identification, and HPLC was used for the content determination. The determination was performed on Agilent ZORBAX Eclipse Plus C18(250 mm× 4.6 mm,5 μ m) column with mobile phase consisted of methano1-water(55:45,V/V) at flow rate of 1.0 m L/min. The detection wavelength was set at 246 nm,and column temperature was 30℃. The sample size was 10 μ L. Results TLC spots of Ficus pandurata Hance were clear and well-separated. The linear range of bergapten were 2.12 ~ 63.6μ g/m L(r = 0.999 9)with average recovery of 99.8%(RSD = 1.12%, n = 6). RSDs of precision, stability and reproducibility tests were lower than 2%.Conclusion The method is simple, accurate and reproducible, and can be used for the quality control of Ficus pandurata Hance.
