基于网络药理学的鸦胆子治疗食管癌的机制研究
Network Pharmacology Study on Mechanism of Fructus Bruceae in Treating Esophageal Carcinoma
【摘要】 目的基于网络药理学探究鸦胆子治疗食管癌可能的作用机制。方法使用中药系统药理学数据库分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)获取鸦胆子有效成分及相关靶点,使用GeneCards、OMIM(Online Mendelian Inheritance in Man,OMIM)获取食管癌相关靶基因,使用Cytoscape 3.7.1软件做出"有效成分-靶点"网络图,使用STRING数据库构建靶蛋白的蛋白互作(protein-protein interaction,PPI)网络图,利用R软件进行基因功能及通路富集分析。结果本研究共筛选出15个鸦胆子的有效成分,并找到了40个与其对应的有效成分靶基因,其中有38个靶基因同时与食管癌相关。通过PPI网络分析最终发现了EGFR、CASP3、CCND1、VEGFA、ERBB2、IL6六个核心基因,其主要富集到与调节肿瘤细胞周期、细胞凋亡、肿瘤内血管生成有关的通路。结论本文初步筛选了鸦胆子治疗食管癌的核心基因,发现了鸦胆子通过多成分、多靶点、多通路协同作用来治疗食管癌的作用机制。
【Abstract】 Objective To explore the possible therapeutic mechanism of Fructus Bruceae for the treatment of esophageal carcinoma by based on network pharmacology. Methods The effective components and related targets of Fructus Bruceae were obtained by using Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP). And then we adopted GeneCards and Online Mendelian Inheritance in Man(OMIM) to obtain esophageal carcinoma related target genes,made the "active ingredient-target" network map by Cytoscape3.7.1,and constructed the protein-protein interaction(PPI)network map of the target protein by using the STRING database, and performed the gene function and pathway enrichment analysis by using R language. Results:A total of 15 active components of Fructus Bruceae were screened out,and 40 target genes corresponding to the active components were obtained,of which 38 target genes were also related to esophageal cancer. Six core genes of EGFR, CASP3, CCND1, VEGFA, ERBB2, and IL-6 were presented after PPI network analysis. Target genes were mainly enriched in the pathways related to the regulation of tumor cell cycle,apoptosis,and angiogenesis in the tumors. Conclusion The article preliminarily screened out the core genes of Fructus Bruceae for the treatment of esophageal carcinoma,and revealed the therapeutic mechanism of Fructus Bruceae in treating esophageal carcinoma through the multi-component, multi-target and multi-pathway synergistic actions.
