MEG3对结肠癌细胞生物活性及VEGF水平影响

【摘要】 目的探讨母系表达基因3(Maternal expression gene 3,MEG3)对结肠癌细胞生物活性及VEGF水平影响。方法将人结肠癌细胞系SW480分为对照组(SW480细胞)、NC组(SW480细胞转染MEG3-NC)和MEG3组(SW480细胞转染MEG3mimics),采用MTT检测SW480细胞活性抑制率,Transwell小室检测侵袭,Hoechst法检测凋亡,免疫印迹检测VEGF蛋白及免疫组化检测VEGF阳性率。结果对照组和NC组SW480细胞lncRNA MEG3水平比较无意义(P>0.05),MEG3组表达升高与其余组别存在意义(P<0.05);不同时间点,对照组及NC组SW480抑制率水平相似(P>0.05),与MEG3组比较,对照组及NC组SW480抑制率均降低(P<0.05),相同时间点,MEG3组SW480抑制率均对照组及NC组(P<0.05),对照组及NC组SW480抑制率水平相似(P>0.05);对照组,NC组及MEG3组SW480细胞侵袭数目分别为(48.90±7.71)个、(44.12±6.90)个及(22.30±3.26)个,对照组、NC组细胞侵袭数目相似(P>0.05),MEG3组细胞侵袭数目低于其他两组(P<0.05);对照组、NC组及MEG3组SW480细胞凋亡率分别为(7.46±0.72)%、(8.12±1.03)%及(31.13±3.22)%,对照组、NC组细胞凋亡率相似(P>0.05);与对照组、NC组比较,MEG3组细胞凋亡率升高(P<0.05);对照组、NC组及MEG3组SW480中VEGF蛋白相对表达水平分别为1.00±0.00、0.91±0.04及0.48±0.05,对照组、NC组VEGF蛋白表达相似(P>0.05);与对照组、NC组比较,MEG3组VEGF蛋白相对表达水平降低(P<0.05);对照组、NC组及MEG3组SW480中VEGF阳性表达不同,对照组、NC组SW480中VEGF阳性表达相似(P>0.05),MEG3组中VEGF阳性表达低于对照组、NC组(P<0.05)。结论在结肠癌细胞中转染MEG3能够抑制活性,减少增殖及加快凋亡,可能与抑制VEGF水平相关。

【Abstract】 Objective To explore the effects of MEG3 on the biological activity of colon cancer cells and the level of VEGF.Methods The human colon cancer cell line SW480 was divided into control group(SW480 cells),NC group(SW480 cells transfected with MEG3-NC) and MEG3 group(SW480 cells transfected with MEG3 mimics).MTT was used to detect the inhibition rate of SW480 cell activity.Cell invasion was detected via transwell and cell apoptosis was measured by Hoechst method.Western blot detection of VEGF protein expression was detected by Western blotting and immunohistochemistry staining.Results The relative amounts of lncRNA MEG3 in SW480 cells in the control group and NC group were 1.00±0.00 and 1.05±0.04,respectively,in which no significant difference between these two groups was shown.The increase in MEG3 protein expression was higher than that in control and NC groups.At the time point,the SW480 inhibition rate in control group and the NC group was similar(P>0.05).Compared with the MEG3 group,the SW480 inhibition rate of the control group and the NC group was reduced.At the same time point,the SW480 inhibition rate in MEG3 group was higher than that in control group and NC group,whereas there was no significant difference between control group and NC group.The number of SW480 cell invasion in the control group,NC group and MEG3 group was(48.90±7.71),(44.12±6.90) and(22.30±3.26),in which the number of cell invasion in MEG3 group was lower than that in the other two groups.The apoptosis rates of SW480 cells in control,NC and MEG3 groups were(7.46±0.72)%,(8.12±1.03)% and(31.13±3.22)%,respectively.Compared with the control and NC groups,the apoptosis rate in MEG3 group was increased.The relative expression level of VEGF protein in control,NC and MEG3 groups were 1.00±0.00,0.91±0.04 and 0.48±0.05,respectively.Compared with control and NC groups,the relative expression level of VEGF protein in MEG3 group was decreased.Moreover,the positive expression of VEGF in MEG3 group was lower than that in control and NC groups.Conclusion Transfection of MEG3 in colon cancer cells could inhibit activity,reduce proliferation and accelerate apoptosis,which might be related to the inhibition of VEGF levels.
