白及属药用植物抗菌作用的谱效关系研究
Relationship between HPLC Fingerprint Chromatogram and Antibacterial Activity of Medicinal Herb of Bletilla Rchb. F.
【摘要】 目的研究白及属药用植物高效液相色谱(HPLC)指纹图谱与其抗菌作用的谱效关系,揭示其抗菌活性的物质基础。方法利用HPLC法建立5批白及、3批黄花白及和4批小白及指纹图谱,采用相似度分析(SA)和聚类分析(HCA)进行评价;以金黄色葡萄球菌、枯草芽孢杆菌为受试菌,以1/最小抑菌浓度(MIC)为指标,运用偏最小二乘回归分析法(PLSR)对共有峰峰面积与1/MIC进行相关性分析,探索其抗菌作用的物质基础,并采用HPLC-四级杆-飞行时间高分辨质谱(HPLC-Q-TOF-HRMS)进行初步鉴定。结果 12批样品指纹图谱共匹配20个共有峰,相似度和聚类分析均可将12批样品分为3类。PLSR分析和HPLC-Q-TOF-MS鉴定结果显示,色谱峰P13(4-甲氧基菲-2,7-二醇)、P12(3,5-二羟基-1-甲氧基-9,10二氢菲)、P17(blestriarene B)、P18(blestriarene C)是抑制金黄色葡萄球菌的主要成分;P12、P18、P19[1-(4-羟基苄基)-4,7-二甲氧基-9,10-二氢菲-2-醇]为抑制枯草芽孢杆菌的主要成分。所有对抑菌作用呈正相关的色谱峰,除P1、P3外,均属菲类和联苄类化合物。结论菲类和联苄类化合物可能是白及属药用植物抑菌作用的主要药效物质基础。
【Abstract】 Objective A spectrum-effect relationship between HPLC fingerprint chromatogram and antibacterial activity of Bletilla Rchb. F. herbs was developed to reveal the material basis of antibacterial activity. Methods Fingerprints of five batches of B. striata(Thunb.)Reiehb. f.,three batches of B. ochracea Schltr.,and four batches of B. formosana(Hayata) Schltr. were established by means of HPLC and further analyzed by SA and HCA.Staphyloccus aureus(STA)and Bacillus subtilis(BS)were used as test bacterium. Partial least-squares regression was employed to investigate the relationship between the common peaks and the 1/MIC to clarify the antibacterial material basis. HPLC-Q-TOF-MS method was used for tentative identification of the major active compounds. Results Twenty common peaks were found in twelve tested samples. SA and HCA performed clearly differentiated and classified the tested samples into three categories. The result indicated P12(coelonin),P13(4-methoxyphenanthrene-2,7-diol),P17(blestriarene B)and P18(blestriarene C)were the main compounds which inhibiting STA;P19(1-(4-hydroxybenzyl)-4,7-dimethoxy-9,10-dihydrophenan-threne-2-ol),P12 and P18 were the main compounds which inhibiting BA. All peaks,except P1 and P3,had positive effect on antibacterial activity belong to bibenzyls and phenanthrenes compounds. Conclusion Phenanthrenes and bibenzyls compounds maybe the main antibacterial material basis of Bletilla Rchb. F..
