miR-361在小鼠哮喘模型中的表达及作用

【摘要】 目的探讨miR-361在小鼠哮喘模型中的作用机制。方法将BALB/c小鼠随机分为正常对照组、哮喘组(卵清蛋白处理组)、治疗组(miR-361治疗组),每组8只,使用实时PCR的方法分析miR-361的表达情况。通过MIRDB软件预测分析miR-361可能结合的蛋白,分析miR-361和转化生长因子-β1(TGF-β1)的表达相关性。荧光素酶报告基因实验和蛋白质免疫印迹法实验检测miR-361对TGF-β1的调节作用。苏木精-伊红染色法(HE染色)分析miR-361对哮喘小鼠肺组织的影响。结果哮喘组中miR-361表达显著下调。miR-361与TGF-β1的3’UTR区域具有结合位点,miR-361与TGF-β1的表达水平负相关。miR-361能够抑制TGF-β1的活性和蛋白表达。miR-361治疗可以缓解哮喘模型肺组织中炎症、黏膜水肿和上皮损伤的发生。结论 miR-361能够通过抑制TGF-β1对卵清蛋白诱导的哮喘小鼠发挥治疗作用。

【Abstract】 Objective To explore the mechanism of miR-361 in mouse asthma model. Methods BALB/c mice were randomly assigned into a normal control group, an asthma group(ovalbumin treatment group) and a treatment group(miR-361 treatment group), with 8 mice in each group. The expression of miR-361 was analyzed by real-time PCR. MIRDB software was used to predict and analyze the possible binding proteins of miR-361, and to analyze the expression correlation between miR-361 and transforming growth factor-β1(TGF-β1). The regulation of miR-361 on TGF-β1 was detected by luciferase reporter gene and Western blot. Hematoxylin-eosin staining(HE staining) was used to analyze the effect of miR-361 on the lung tissue of asthmatic mice. Results The expression of miR-361 was significantly decreased in asthma group. MiR-361 had a binding site with the 3’UTR region of TGF-β1, and miR-361 had a negative correlation with the expression level of TGF-β1. MiR-361 could inhibit the activity and protein expression of TGF-β1. MiR-361 treatment could alleviate the inflammation, mucosal edema and epithelial damage in the lung tissue of asthma model. Conclusion MiR-361 has therapeutic effect on ovalbumin induced asthmatic mice by inhibiting TGF-β1.
