响应面法优化桃儿七鬼臼毒素的超声提取工艺

【摘要】 目的:采用超声波辅助提取法,优化桃儿七中鬼臼毒素的提取工艺,比较不同地区野生桃儿七中鬼臼毒素的含量。方法:以桃儿七中鬼臼毒素的含量为评价标准,设计单因素和响应面试验,考察料液比、超声时间、超声温度及甲醇体积分数4个因素对桃儿七鬼臼毒素提取效果的影响。结果:优化最佳提取工艺条件为:料液比为1∶40.3 (g∶mL),超声时间为20.86 min,超声温度为55.74℃,甲醇体积分数为81.99%。结论:对不同地区的桃儿七干燥根中鬼臼毒素的含量进行测定,发现西藏地区桃儿七中鬼臼毒素的含量最高为4.18%,从而为桃儿七中鬼臼毒素的开发利用提供科学依据。

【Abstract】 Objective: Ultrasound-assisted extraction method was used to optimize the extraction process of podophyllotoxin(PPT) from Sinopodophyllum hexandrum and to compare the content of PPT from wild S. hexandrum from different areas. Methods: The content of PPT in S. hexandrum was used as the evaluation standard, and the single factor and response surface tests were designed to investigate the effects of four factors(solid-liquid ratio, ultrasonic time, ultrasonic temperature and methanol volume fraction) on the extraction of PPT. Results: The optimal extraction conditions were as follows: the solid-liquid ratio was 1∶40.3(g ∶mL), the ultrasonic time was 20.86 min, the ultrasonic temperature was 55.74℃, and the methanol volume fraction was 81.99%. Conclusion: The PPT content in the dried root of S. hexandrum from different areas was determined. The highest PPT content in S. hexandrum in Tibet was 4.18%. Thus, this research provides a reference for the development and utilization of the PPT in S. hexandrum.
