基于网络药理学探究心达康片治疗冠心病心绞痛的作用机制

【摘要】 目的基于网络药理学探究心达康片治疗冠心病心绞痛的作用机制,为临床提供理论依据。方法挖掘文献获取心达康片的有效成分,利用中医药百科全书(ETCM)数据库和人类基因数据库(Gene Cards)获取疗效靶标,使用KOBAS 3.0服务器对靶标进行KEGG通路富集分析,构建"药物-靶标-通路"网络。利用STRING数据库进行对疗效靶标进行蛋白相互作用分析,并利用网络分析工具获取核心靶标。结果心达康片的有效成分为槲皮素、异鼠李素、山柰酚,疗效靶标共490个,其中159个靶标富集在脂质与脂蛋白代谢、PI3酶(PI3K)/丝氨酶/苏氨酸激酶(Akt)信号通路、血小板激活与聚集、HIF-1信号通路和Rap1信号通路这5条重要通路。5个核心靶标分别为INS、AKT1、MAPK3、GAPDH、IL-6。结论心达康片通过多成分、多靶标、多通路治疗冠心病心绞痛,通过抑制炎症、氧化损伤和细胞凋亡,减少冠脉痉挛,减轻冠脉狭窄,改善血供发挥疗效。

【Abstract】 Objective:To explore the mechanism of Xindakang Tablets(XDK)on coronary disease angina pectoris based on network pharmacology and to provide theoretical basis for clinical application. Methods:The effective components of XDK were obtained by literature mining,and the therapeutic targets were obtained by ETCM database and Genecards database. KOBAS 3.0 server was used to make a KEGG pathway enrichment analysis of the target,and the "drug-target-pathway" network was constructed. STRING database was used to analyze protein-protein interaction of therapeutic targets,network analysis tools to obtain core targets. Results:The active components of XDK were quercetin,isorhamnetin and kaempferol. There were 490 therapeutic targets,among which 159 targets were concentrated in five important pathways,such as lipid and lipoprotein metabolism,PI3 K-Akt signaling pathway,platelet activation and aggregation,HIF-1 signaling pathway and Rap1 signaling pathway. Five core targets includes INS,AKT1,MAPK3,GAPDH and IL-6. Conclusion:XDK can treat angina pectoris of coronary heart disease through multi-component,multi-target and multi-channel. It can reduce coronary spasm,reduce coronary stenosis and improve blood supply by inhibiting inflammation,oxidative damage and apoptosis.
