射干麻黄汤对急性呼吸衰竭模型大鼠的干预效果及作用机制研究
Intervention Effect and Mechanism of Shegan Mahuang Decoction on ARF Model Rats
【摘要】 目的观察射干麻黄汤对急性呼吸衰竭模型大鼠的干预效果。方法选取40只SD雄性大鼠,10只为正常组,其余30只大鼠建立急性呼吸衰竭模型,并分为模型组、西药组、射干麻黄汤组,正常组、模型组大鼠使用0.9%氯化钠注射液进行灌胃处理,西药组大鼠使用硫酸阿托品注射液干预,射干麻黄汤组大鼠使用射干麻黄汤进行灌胃处理。对各组大鼠呼吸频率及p H、二氧化碳分压(PaCO2)、氧分压(PaO2)水平进行检测,酶联免疫吸附实验法检测白细胞介素-8(IL-8)、白细胞介素-17(IL-17)、超敏C反应蛋白(hs-CRP)水平,免疫透射比浊法检测谷胱甘肽过氧化物酶(GSH-Px)、过氧化脂(LPO)、丙二醛(MDA)水平;Western blotting法检测Bcl-2、Bax、Caspase3表达。结果模型组、西药组、射干麻黄汤组大鼠呼吸频率、pH、PaCO2、PaO2水平低于正常组,射干麻黄汤组大鼠呼吸频率、pH、PaCO2、PaO2水平高于西药组、模型组(P <0.05)。模型组、西药组、射干麻黄汤组大鼠IL-8、IL-17、hs-CRP、LPO、MDA水平、Bcl-2、Caspase3表达高于正常组,GSH-Px水平、Bax表达低于正常组(P <0.05);西药组、射干麻黄汤组大鼠IL-8、IL-17、hs-CRP、LPO、MDA水平、Bcl-2、Caspase3表达低于模型组,GSH-Px水平、Bax表达高于模型组,且射干麻黄汤组大鼠IL-8、IL-17、hs-CRP、LPO、MDA水平、Bcl-2、Caspase3表达低于西药组,GSH-Px水平、Bax表达高于西药组(P <0.05)。结论使用射干麻黄汤对急性呼吸衰竭大鼠进行干预,能够改善大鼠呼吸频次及血气水平,减轻大鼠炎症反应及氧化应激损伤,抑制肺组织细胞凋亡。
【Abstract】 Objective:To explore the intervention effect and mechanism of Shegan Mahuang Decoction on acute respiratory failure(ARF)model rats. Methods:10 of 40 male SD rats were the normal group;the rest 30 rats were divided into the model group,Western medicine(WM)group,Shegan Mahuang Decoctio(SMD)group. The normal group and the model group were treated with normal saline,WM group with atropine sulfate injection,SMD group with Shegan Mahuang Decoction. The respiratory rate,p H,PaCO2 and PaO2 of each group were detected.The levels of IL-8,IL-17,hs-CRP,GSH-Px,LPO and MDA were detected by ELISA. The expression of Bcl-2,Bax and caspase-3 was detected by Western blotting. Results:The levels of respiratory frequency,pH,PaCO2 and PaO2 in the model group,WM group and SMD group were lower than those in the normal group,while those in SMD group were higher than those in WM group and the model group(P < 0.05). The expression of IL-8,IL-17,hs CRP,LPO,MDA,Bcl-2 and Caspase-3 in the model group,WM group and SMD group was higher than that in the normal group,and the expression of GSH PX and Bax was lower than that in the normal group(P < 0.05);the expression of IL-8,IL-17,hs CRP,LPO,MDA,Bcl-2 and Caspase-3 in WM group and SMD group were lower than that in the model group,and the expression of GSH-PX and Bax was higher than that in the model group.The expression of IL-8,IL-17,hs-CRP,LPO,MDA,Bcl-2,Caspase-3,GSH PX and Bax in SMD group was lower than that in WM group(P < 0.05). Conclusion:The intervention of Shegan Mahuang Decoction on rats with acute respiratory failure can improve the respiratory frequency and blood gas level,reduce the inflammatory response and oxidative stress injury,and inhibit the apoptosis of lung tissue cells.
