翻白草对TNBS诱导的急性溃疡性结肠炎大鼠促炎因子及趋化因子的影响
Effects of Potentilla Discolor Bunge on proinflammatory and chemokines in TNBS induced acute ulcerative colitis rats
【摘要】 目的:研究翻白草对三硝基苯磺酸(TNBS)诱导的实验性溃疡性结肠炎大鼠的治疗作用,并阐明其对促炎因子IL-17、TNF-α、IFN-γ、MIP-1α以及趋化因子(RANTES)表达的影响。方法:采用TNBS-乙醇灌肠法复制溃疡性结肠炎大鼠模型,实验分为空白对照组、模型对照组、柳氮磺吡啶组(阳性药)、翻白草醇提物高、中、低剂量组6组,连续给药14 d后取结肠组织进行疾病活动指数(disease activity index,DAI)和苏木素-伊红染色(HE)评价,通过蛋白芯片检测大鼠结肠组织中IL-17、TNF-α、IFN-γ、MIP-1α以及趋化因子(RANTES)含量。结果:翻白草各剂量组DAI水平较模型组均明显下降,其中翻白草高、中剂量组能够明显降低大鼠结肠组织炎细胞浸润的程度,经翻白草干预后大鼠结肠组织中促炎因子IL-17、TNF-α、IFN-γ、MIP-1α及RANTES表达减少。结论:翻白草醇提物通过调节炎症因子表达,抑制趋化因子募集粒细胞浸润,有效改善TNBS诱导的急性溃疡性结肠炎大鼠的肠黏膜损伤。
【Abstract】 Objective:To investigate the therapeutic effects of Potentilla Discolor(PD) on trinitrobenzene sulfonic acid(TNBS)-induced experimental ulcerative colitis in rats and to elucidate its effects on the expression of proinflammatory cytokines IL-17, TNF-α, IFN-γ, MIP-1α, and chemokines(RANTES). Method:The rat model of ulcerative colitis was induced by TNBS-ethanol enema. The rats were divided into 6 groups: blank control group, model control group, sulfasalazine group(positive drug), and high, middle and low dose groups of ethanol extract of PD. After 14 days of continuous administration, colon tissues were harvested and evaluated for disease activity index(DAI) and hematoxylin-eosin staining(HE), and IL-17, TNF-α, IFN-γ, MIP-1α, as well as chemokine(RANTES) contents in rat colon tissues were detected by protein microarray. Result:The level of DAI in PD group was significantly lower than that in model group, and the level of inflammatory cell infiltration in colon tissue of rats was significantly decreased in PD high and medium dose groups, and the expression of proinflammatory cytokines IL-17, TNF-α, IFN-γ, MIP-1α and RANTES in colon tissue of rats was decreased after PD intervention. Conclusions: Ethanol extract of PD effectively ameliorated the state of TNBS-induced acute ulcerative colitis by regulating the expression of inflammatory factors and inhibiting the recruitment of granulocyte infiltration by chemokines.
