柴胡皂苷a抑制PTZ诱导小鼠海马星形胶质细胞增殖和多药转运蛋白的表达
Saikosaponin a inhibits PTZ induced proliferation and expression of multidrug transporter in mouse hippocampal astrocytes
【摘要】 目的:探究柴胡皂苷a(SSa)对戊四氮(PTZ)诱导小鼠海马星形胶质细胞增殖及多药转运蛋白表达的影响。方法:体外培养小鼠海马星形胶质细胞,按随机数字表法分为对照组(A组)、10 mmol/L PTZ刺激2 h组(B组)、10 mmol/L PTZ刺激前采用0.625 mg/L或1.25 mg/L SSa药物处理6 h(C和D)组。采用MTT检测细胞增殖活性,Realtime PCR和Western blot分别检测多药转运蛋白P-gp、MRP2、BCRP和腺苷激酶(ADK)mRNA和蛋白表达水平。结果:与A组比较,PTZ处理后B组细胞增殖率明显升高,且P-gp、MRP2、BCRP和ADK的mRNA和蛋白表达水平均显著升高;与B组比较,C和D组细胞增殖率均显著降低,P-gp、MRP2、BCRP和ADK的mRNA和蛋白表达水平均显著下调,且随着SSa浓度升高,D组的作用效果更明显。结论:柴胡皂苷(SSa)可有效抑制PTZ诱导的海马星形胶质细胞增殖,同时降低细胞中多种多药转运蛋白以及ADK的表达。
【Abstract】 Objective:To study effects of Saikosaponin a(SSa) on cell proliferation and multidrug transporter proteins expression in mouse hippocampal astrocytes induced by pentylenetetrazol(PTZ). Methods:Mouse hippocampal astrocytes were cultured in vitro and randomly divided into control groups(group A), 10 mmol/L PTZ-induced for 2 h group(group B), 0.625 or 1.25 mg/L SSa treatment for 6 h group before treating with PTZ(group C and D). The cell proliferation were detected by MTT assay; the mRNA and protein expression of multidrug transporter proteins(P-gp, MRP2 and BCRP) and adenosine kinase(ADK) were assessed by Realtime PCR and western blot. Results:Compared with group A, the cell proliferative rate significantly rised in group B, meanwhile the mRNA and protein expression of P-gp, MRP2, BCRP and ADK markedly increased. Compared with group B, the cell proliferative rate significently reduced in groups C and D, meanwhile the mRNA and protein expression of P-gp, MRP2, BCRP and ADK markedly decreased. Additionally, with the increased dosage of SSa, the effects in group D were more obvious. Conclusion:Saikosaponin a(SSa) can effectively inhibit PTZ-induced proliferation of hippocampal astrocytes and also reduce the expression of multiple multidrug transporter proteins and ADK in hippocampal astrocytes.
