青霉菌灭活菌丝体提取物对烟草BY-2细胞抗病防卫反应的诱导作用

【摘要】 青霉菌灭活菌丝体水提取物(water extract of dry mycelium of Penicillium chrysogenum, DMP)可以作为激发子诱导植物抗病防卫反应。本研究对DMP中多糖(PsD)和多肽(PDMP)激发子进行分离提取,并以chitin和Flg22为参照,研究Ps D和PDMP对烟草BY-2细胞抗病防卫反应的诱导作用。结果表明:PsD和PDMP能够诱导BY-2细胞活性氧迸发,分别在处理后5和25 min达到峰值。PsD和PDMP能够诱导烟草BY-2细胞苯丙烷途径关键基因PAL、4CL和C4H和几丁质基因NtCHitIV的表达,并积累酚类次生代谢产物。基础免疫标志基因Pti5表达量在Ps D和PDMP诱导下显著升高,说明Ps D和PDMP与植物细胞互作的机理可能为模式识别受体介导的MAMP识别,并诱导下游抗病防卫反应。以上结果表明, PsD和PDMP能够激活烟草BY-2细胞基础抗性,诱导抗性防卫反应。

【Abstract】 Water extract of dry mycelium of Penicillium chrysogenum(DMP) serves as an elicitor to induce plant defense responses. In this study, the polysaccharide(PsD) and polypeptide(PDMP) were isolated from DMP, and the effects of PsD and PDMP on defense responses in tobacco BY-2 cells were studied, using chitin and Flg22 as references. The results showed, PsD and PDMP induced oxidative burst in tobacco BY-2 cells, the peaks were observed 5 min after PsD treatment, and 25 min after PDMP treatment, respectively. Transcripts of key regulators genes of phenylpropanoid pathway, PAL, 4CL, and C4H, as well as a chitinase gene NtCHitIV, were rapidly induced both in PsD and PDMP treated cells. Transcripts of marker gene of MAMP/PAMP-triggered immunity(PTI) Pti5 was rapidly induced both in PsD and PDMP treated cells, indicating that PsD and PDMP actor as MAMP to trigger PTI, and activate following defense responses. This study suggests that PsD and PDMP act as elicitors and activate defense responses though PTI in tobacco BY-2 cell.
