ISSR标记技术辅助形态学的红曲菌株分类鉴定

【摘要】 以市售红曲产品中分离的17株红曲菌株和8株标准菌株为主要材料,根据它们的菌落形态和显微形态特征进行分类,25株菌株归为6类。同时,运用内部简单重复序列(ISSR)标记分类法进行聚类分析,在相似性水平为0.9时,25株菌株聚为5类。综合形态学分类鉴定法和ISSR标记分类法结果,25株红曲菌株被聚为3大类,分别是红色红曲菌(Monascus ruber)、紫色红曲菌(Monascus purpureus)和血红红曲菌(Monascus sanguineus)。结果表明,传统形态学分类方法结合ISSR标记分类法能为红曲菌株的分类鉴定提供综合参考信息。也可为了解市售红曲相关发酵产品生产菌株的分类与特征、进一步发掘和保护红曲菌株资源提供参考。

【Abstract】 Monascus strains, including 17 isolates from different commercial Hongqu products and 8 standard strains, were classified into 6 groups based on their colonial and microscopic morphologies. In addition, inter-simple sequence repeat(ISSR) markers was applied to access the genetic diversity of these 25 of Monascus strains. All the tested strains were grouped into 5 clusters at a 0.9 similarity level. By integrated morphological identification and ISSR classification results, 25 tested strains were clustered into 3 groups, which were Monascus ruber, Monascus purpureus and Monascus sanguineus,respectively. It was suggested that combination of morphological classification methods with the ISSR method could provide a comprehensive information for classification and identification of Monascus strains. It was also provided information for learning the strain characteristics of Monascus-related fermentation products for exploring and protecting the biological resources of Monascus strains.
