非小细胞肺癌患者血浆中lncRNA-MALAT1和lncRNA-ATB的表达及诊断价值
Diagnostic value of plasma IncRNA-MALAT1 and lncRNA-ATB levels in non-small cell lung cancer
【摘要】 目的:评价血浆长链非编码RNA肺腺癌转移相关转录因子1 (metastasis-associated lung adenocarcinoma transcript 1,lncRNA-MALAT1)及转化生长因子β激活的长链非编码RNA(lncRNA activated by TGF-β,lncRNAATB)诊断非小细胞肺癌(non-small cell lung cancer,NSCLC)的临床价值。方法:收集60例NSCLC患者和60例良性肺病患者的血浆标本,采用荧光定量PCR技术测定lncRNA-MALAT1及lncRNA-ATB的相对表达水平,分析其与NSCLC患者临床病理特征的关系。分别构建血浆lncRNA-MALAT1及lncRNA-ATB诊断NSCLC的ROC曲线,评估其诊断效果,并与血清癌胚抗原及细胞角蛋白十九片段(Cyfra21-1)进行比较。结果:与良性肺病患者相比,NSCLC患者血浆lncRNA-MALAT1表达明显下调,lncRNA-ATB表达明显上调(P<0.05)。血浆lncRNA-MALAT1及lncRNA-ATB表达水平与NSCLC患者性别、年龄、吸烟指数、组织病理类型及TNM分期等无显著相关(P均>0.05)。血浆lncRNA-MALAT1和lncRNA-ATB诊断NSCLC的ROC曲线下面积均明显高于血清癌胚抗原及Cyfra21-1;二者诊断NSCLC的准确性高于血清癌胚抗原及Cyfra21-1,二者联合检测准确性较单独检测高。结论:血浆lncRNAMALAT1及lncRNA-ATB可作为诊断NSCLC的生物标志物。
【Abstract】 Objective:To evaluate the clinical value of plasma IncRNA-metastasis-associated lung adenocarcinoma transcript 1(MALAT1) and IncRNA-activated by TGF-β(ATB) in the diagnosis of nonsmall cell lung cancer(NSCLC).Methods:Quantitative real-time PCR was used to detect the expression level of IncRNA-MALAT1 and IncRNA-ATB in plasma samples of 60 patients with NSCLC and 60 patients with benign lung disease.The potential correlations between plasma IncRNA-MALAT1 and lncRNA-ATB levels and clinical pathological characteristics in NSCLC patients were analyzed.Receiver operating characteristic curves(ROC) was plotted by statistical software,and area under the curve(AUC)was used to assess the performance of plasma IncRNA-MALAT1 and IncRNA-ATB in diagnosis of NSCLC.These two performances then were compared with those of serum carcinoembryonic antigen(CEA) and cytokeratin-19-fragment(Cyfra21-1).Results:Compared to benign lung disease patients,the plasma levels of IncRNA-MALAT1 was greatly decreased(P <0.05) and lncRNA-ATB was increased significantly(P <0.05) in NSCLC patients.The plasma levels of lncRNA-MALAT1 and IncRNA-ATB in NSCLC patients were not correlated with gender,age,smoking index,histopathological type and tumor TNM stage(P>0.05).The ROC curves illustrated that AUCs of lncRNA-MALAT1 and IncRNA-ATB was higher than those of CEA and Cyfra21-1.Moreover,the combination of lncRNA-MALAT1 and lncRNA-ATB yielded a higher accuracy as compared to lncRNA-MALAT1 or lncRNA-ATB alone.Conclusion:Plasma lncRNA-MALAT1 and lncRNA-ATB could be used as biomarkers for diagnosis of NSCLC.
