桑寄生提取物对人结肠癌HT-29细胞侵袭迁移的影响及作用机制
Mechanism and effect of water extract from herba taxillion the invasion and migration of human colorectal cancer HT-29 cells
【摘要】 目的探讨桑寄生提取物对人结肠癌HT-29细胞侵袭迁移的影响及作用机制。方法不同浓度(0、0.5、1、2、4、8g/ml)的桑寄生提取物处理HT-29细胞24、48或72h后,分别采用细胞计数试剂盒法、侵袭小室法、划痕实验、免疫印迹试验检测不同浓度桑寄生提取物对HT-29细胞增殖、侵袭、迁移能力以及细胞侵袭迁移相关蛋白、PI3K/AKT信号通路相关蛋白的影响。结果桑寄生提取物可明显抑制HT-29细胞增殖,呈浓度和时间依赖性;与空白对照组(0g/ml)相比,桑寄生提取物能降低HT-29细胞侵袭、迁移能力,呈剂量依赖性;能下调细胞侵袭迁移相关蛋白[基质金属蛋白酶2(MMP-2)、MMP-9、血管内皮生长因子(VEGF)]及p-AKT、p-PI3K蛋白表达。结论桑寄生提取物能抑制人结肠癌HT-29细胞的增殖、侵袭及迁移,其作用机制可能与抑制PI3K/AKT信号通路有关。
【Abstract】 Objective To investigate the mechanism and effects of water extract from herba taxilli(WHT) on the invasion and migration of human colorectal cancer HT-29 cells. Methods HT-29 was treatment with WHT on different concentration(0, 0.5, 1, 2, 4,8 g/ml) for 24, 48 or 72 h. CCK-8 assay, transwell, tarch test and western blot was used to detect the effect of WHT on cell proliferation,invasion, metastasis, expression of cell invasion and migration related proteins and PI3 K/AKT signaling pathway protein, respectively. Results WHT inhibited the cell proliferation of human colorectal cancer HT-29 cells in a dose-and time-dependent manner. Compared with the blank group, WHT inhibited the invasion and metastasis of HT-29 in a dose-dependent manner. WHT down-regulated protein expressions of MMP2, MMP9 VEGF and p-AKT and p-PI3 K. Conclusion WHT inhibits the invasion and migration of human colorectal cancer HT-29 cells, the mechanism might be by inhibiting PI3 K/AKT signaling pathway.
