黄芪生脉饮中多糖水解单糖指纹图谱的研究
Research on HPLC fingerprint of monosaccharide from hydrolyzed polysaccharide of Huangqi Shengmai decoction
【摘要】 为了建立黄芪生脉饮(Huangqi Shengmai decoction,HSD)中多糖水解单糖的高效液相色谱指纹图谱,对其质量标准的建立提供理论支持,用三氟乙酸(Trifluoroacetic acid, TFA)对HSD多糖提取物进行水解和PMP(1-phenyl-3-methy-5-pyrazolone)衍生化处理,建立高效液相色谱-紫外法(HPLC-UV)测定多糖水解衍生物,并对其进行方法学考察。采用"中药指纹图谱相似度评价系统"软件,评价分析10个不同批次的HSD中多糖水解单糖的相似度。得到最佳TFA水解浓度为2 mol/L,总多糖样品5.0 mg时,PMP衍生化条件为PMP用量400μL,反应时间80 min。建立的色谱指纹图谱精密度、重复性和稳定性的RSD值均小于5%,各批次的相似度均在0.990以上,符合指纹图谱测定要求,可以用于HSD中多糖水解单糖质量的评价。
【Abstract】 To obtain a HPLC fingerprint of monosaccharide from hydrolyzed polysaccharide in Huangqi Shengmai decoction(HSD), which can be theoretical basis for establishing quality standard of HSD,hydrolysisof polysaccharide in HSD by trifluoroacetic acid(TFA)followed by 1-phenyl-3-methy-5-pyrazolone(PMP) derivation was performed. An HPLC-UV fingerprint method was established to analyze the derivatives from the hydrolysis of polysaccharide. Similarity evaluation system for chromatography fingerprint of TCM was used to evaluate the similarity of monosaccharide derivatives from hydrolyzed polysaccharide in ten batches.The optimal concentration of TFA to hydrolyze polysaccharide is 2 mol/L. When the amount of HSD is 5.0 mg, the dose of PMP is 400 μL and the reaction time is 80 min. The RSD values of precision, inter-day repeatability and stability of the established HPLC fingerprint are lower than 5%, and the similarity of each batch is higher than 0.990, which meets the requirements of fingerprint and can be used to evaluate the quality of monosaccharide from hydrolyzed polysaccharides in HSD.
