穿龙薯蓣茎段组培体系的建立
Establishment of Tissue Culture System of Dioscorea nipponica Stem Segment
【摘要】 为了探明不同处理对穿龙薯蓣茎段组织培养的影响,建立穿龙薯蓣茎段快速繁殖体系,为其规模化种植提供充足的材料。以穿龙薯蓣幼嫩带节茎段作为外植体,研究了消毒时间及不同培养基激素配比对穿龙薯蓣腋芽诱导、增殖分化及生根的影响。结果表明,MS+0.2 mg·L-1 NAA+2.0 mg·L-1 6-BA为腋芽诱导的适宜培养基,诱导率为86.67%;MS+0.2 mg·L-1 NAA+3.0 mg·L-1 6-BA较适宜穿龙薯蓣增殖分化,增殖倍数达4.58;1/2 MS+0.1 mg·L-1 NAA处理下穿龙薯蓣试管苗生根效果好,生根率达73.34%;以蛭石∶腐质土∶河沙=1∶1∶1为移栽基质,移栽成活率可达63.20%。研究表明,不同激素组合对穿龙薯蓣再生体系的建立有不同效果。
【Abstract】 In order to investigate the effects of different treatments on tissue culture of Dioscorea nipponica stem,a rapid propagation system was established to provide sufficient materials for large-scale cultivation.Also,the effects of disinfection time and different hormone ratios in different media on the induction,proliferation,differentiation and rooting of axillary buds of D. nipponica were studied.The results showed that MS + 0.2 mg·L-1 NAA + 2.0 mg·L-1 6-BA was the appropriate medium for axillary bud induction with 86.67% induction rate,while MS + 0.2 mg·L-1 NAA + 3.0 mg·L-1 6-BA was suitable for the proliferation and differentiation of D. nipponica with the proliferation multiple being 4.58.In addition,the rooting effect of D. nipponica seedlings treated with 1/2 MS+0.1 mg·L-1 NAA was the best,and the rooting rate reached 73.34%.Meanwhile,the survival rate of transplanting was over 63.20% with mixed matrix(1 vermiculite∶1 humic soil∶1 river sediment). In conclusion, different hormone combinations had different effects on regeneration system of D. nipponica.
