纳米吲哚菁绿酶响应水凝胶的合成及体外抗肿瘤实验研究

【摘要】 目的:在本研究中,我们合成了一种新型智能载药水凝胶(nano ICG@MMP-gel),检测其光敏特性及体外抗肿瘤效果,以期为舌癌治疗提供新策略。方法:首先利用溶剂-反溶剂法制备纳米吲哚菁绿,并装载至基质金属蛋白酶响应水凝胶内,合成nano ICG@MMP-gel,并用扫描电镜和体外药物释放试验对其进行表征;然后研究nano ICG@MMP-gel的体外光热特性、活性氧产生情况,最后通过MTT法、细胞划痕实验及侵袭实验研究体外抗肿瘤作用。结果:纳米吲哚菁绿在基质金属蛋白酶响应水凝胶中均匀分布,并且在基质金属蛋白酶条件下能缓慢释放。经808nm近红外光照射后,nano ICG@MMP-gel显示出理想的光热特性,可产生大量的活性氧,并且可抑制SCC-15的增殖、侵袭和转移。结论:在808nm近红外照射下,nano ICG@MMP-gel具有理想的光敏特性及良好的抗肿瘤效果,为舌癌治疗提供了理论和实验依据。

【Abstract】 Objective:In this study, we synthesized a novel intelligent drug loaded hydrogel(denoted as nano ICG@MMP-gel), to detect its photosensitivity and in vitro anti-tumor effect, so as to provide a new strategy for the treatment of tongue cancer. Methods: First, nano ICG@ MMP-gel was synthesized and characterized by scanning electron microscopy and drug-release assays. Second, the photosensitivity properties of this drug delivery system were studied by temperature measurements and ROS test. Last, the antitumor efficiency was studied by MTT, scratch assay and invasion test in vitro.Results: Nano indocyanine green was evenly distributed in the matrix metalloproteinase-responding hydrogel, and under the condition of matrix metalloproteinases, nano ICG@MMP-gel could slowly release after being irradiated by 808 nm nearinfrared light. The nano ICG@MMP-gel showed ideal photothermal characteristics, produced a large number of reactive oxygen species, and inhibited the proliferation, invasion and metastasis of SCC-15 Conclusion: The study of nano ICG@MMP-gel with 808 nm NIR irradiation could provide theoretical and experimental basis for the treatment of tongue cancer.
