细香葱种质资源花粉活力及测定方法的比较

【摘要】 花粉活力是杂交育种能否成功的决定因素之一。为了探明香葱不同种质资源的花粉活力状况,采用TTC染色法、联苯胺染色法、离体萌发法进行香葱花粉活力不同测定方法的比较,筛选出适宜简单快速测定香葱花粉活力的方法,并采用该方法对29个香葱种质资源的花粉活力进行检测。结果表明,采用联苯胺染色法对香葱花粉活力进行测定最为快速准确,平均花粉活力为74.04%。采用联苯胺染色法对29份香葱种质进行花粉活力测定,AF14花粉活力最高,为80.20%,其次为AF93和AF94,种质间花粉活力存在显著差异。综合分析表明,联苯胺染色法为香葱花粉活力快速测定的最佳方法,AF14等种质可优先用于材料创制。

【Abstract】 Pollen viability is one of the decisive factors for the success of hybrid breeding. In order to ascertain the pollen viability of different germplasm resources of Allium schoenoprasum,TTC staining method, benzidine staining method,and in vitro germination method were compared,a simple and rapid method to determine pollen viability was screened out.Then this method was used to detect the pollen viability of different germplasm resources.The results showed that in TTC, benzidine and in vitro germination methods, benzidine dyeing was the best method for rapid determination of pollen viability of Allium schoenoprasum,the mean viability was 74.04%. The pollen viability of 29 germplasm was determined by benzidine staining,and the pollen viability of AF14 germplasm was the highest(80.20%), followed by that of AF93 and AF94. There were significant differences in pollen viability among some cultivars. Comprehensive analysis showed that the benzidine staining method is the best method for the rapid determination of Allium schoenoprasum and germplasm such as AF14 could be preferentially used for material creation.
