晚期糖基化终末产物与神经退行性疾病的相关性

【摘要】 随着老龄化程度的日益加剧,我国的神经退行性疾病特别是阿尔茨海默症、帕金森氏症的发病率也相应增加,给人类带来了巨大的疾病负担。神经退行性疾病的发病机制非常复杂,研究证实,晚期糖基化终末产物(AGEs)与神经退行性疾病的发生和进展联系密切。AGEs是由还原糖的羰基与蛋白质、脂类或核酸的游离氨基之间的非酶反应形成的一组化合物,随着年龄的增大,AGEs在体内逐渐聚集。目前越来越多的证据显示,由于蛋白质和多肽之间存在着抗蛋白酶交联,所以AGEs的形成可能会促进与神经退行性疾病有关的几种蛋白质的沉积,从而促进神经退行性疾病的发展。本文综述了近年来AGEs与神经退行性疾病的相关性研究,介绍了AGEs的来源、AGEs与阿尔茨海默症和帕金森氏症这两种重要的神经退行性疾病的关系及降低AGEs的营养饮食方法,为开发新的神经退行性疾病的预防和治疗策略提供重要基础和线索。

【Abstract】 The incidence of neurodegenerative diseases,especially the Alzheimer’s Disease and Parkinson’s Disease has increased rapidly with aging in China,bringing a huge burden of diseases to human beings. The pathogenesis of neurodegenerative diseases is very complex. Studies have confirmed that Advanced Glycation End Products (AGEs) are closely related to the occurrence and progression of neurodegenerative diseases. AGEs are a group of compounds formed by nonenzymatic reactions of the carbonyl group of reducing sugars with proteins,lipids,or free amino acids of nucleic acids. More and more evidences have showed that the formation of AGEs may promote the deposition of several proteins associated with neurodegenerative diseases,thereby promoting the development of neurodegenerative diseases due to the antiprotease cross-linking between proteins and peptides. This article reviewed the recent research on the relationship between AGEs and neurodegenerative diseases,which introduced the sources of AGEs,the relationship between AGEs and Alzheimer’s disease and Parkinson’s disease and nutritional dietary approaches to reduce AGEs,to provide important basis and clues for the development of new strategies for prevention and treatment of neurodegenerative diseases.
