燕麦β葡聚糖对糖尿病大鼠肾功能及其肠道菌群的影响
Effects of Oat β-Glucan on Renal Function and Intestinal Flora in Diabetic Rats
【摘要】 目的:探讨燕麦β葡聚糖(oatβ-glucan,OG)对糖尿病大鼠肾功能及肠道菌群的影响。方法:采用单侧肾切除+链脲佐菌素(65mg/kg·BW)一次性腹腔注射方式构建糖尿病肾病(diabetes nephropathy,DN)大鼠模型,将造模成功32只SD雄性大鼠随机分为4组:模型对照组(蒸馏水灌胃)和低、中、高燕麦β葡聚糖干预组(分别用0. 275、0. 55、1. 1g/kg·BW燕麦β葡聚糖灌胃),每组8只。另取16只大鼠行假手术并随机分组:正常对照组(蒸馏水灌胃)和燕麦β葡聚糖对照组(0. 55g/kg·BW燕麦β葡聚糖灌胃)。饲养8周,在第8周末采集血液、尿液和粪便,进行血糖、肾功能检测,用HE染色进行肾脏形态学观察,16S r DNA检测分析肠道菌群多样性。结果:高剂量燕麦β葡聚糖能显著降低DN大鼠血糖(P <0. 05),低、中剂量燕麦β葡聚糖能显著降低DN大鼠血尿素氮、肌酐;中、高剂量燕麦β葡聚糖能降低DN大鼠的尿酸/肌酐比值(P <0. 05);高剂量燕麦β葡聚糖对DN大鼠的肾小球系膜基质增生和基底膜增厚,肾小管上皮细胞肿胀变形、脱落,系膜细胞增生有显著改善作用;高剂量燕麦β葡聚糖能显著升高DN大鼠肠道菌群丰度和多样性、厚壁菌门/拟杆菌门的比值(P <0. 05)。结论:燕麦β葡聚糖能改善糖尿病大鼠肾功能,增加肠道菌群丰度和多样性及升高厚壁菌门/拟杆菌门的比值,这可能是其延缓DN的机制之一。
【Abstract】 Objective To investigate the effects of oat β-glucan on renal function and intestinal flora in diabetic rats. Method A rat model of diabetic nephropathy( DN) was established by unilateral nephrectomy plus streptozotocin( 65 mg/kg · BW). Totally 32 male Sprague-Dawley rats were randomly selected and divided into 4 groups including model control group( distilled water gavage) and low,medium and high-dose oat β-glucan intervention group( 0. 275 g/kg·BW,0. 55 g/kg·BW and 1. 1 g/kg·BW oat β-glucan respectively) were administered,irrespectively. Another 16 rats underwent sham operation were randomized into normal control group( distilled water gavage)and oat β-glucan control group( 0. 55 g/kg·BW oat β-glucan). The experiment lasted for 8 weeks. At the 8 th week,blood,urine and feces were collected for blood glucose and renal function tests. HE staining was used to observe the morphology of the kidney,and 16 S rDNA was used to analyze the diversity of the intestinal flora. Result High-dose oat β-glucan significantly reduced blood glucose in diabetic nephropathy rats( P < 0. 05). Low-and medium-dose oat β-glucan significantly reduced blood urea nitrogen and creatinine in rats with diabetic nephropathy. Medium-and high-dose oat β-glucan could reduce the uric acid/creatinine ratio in diabetic nephropathy rats( P < 0. 05).High-dose oat β-glucan could improve glomerular mesangial matrix hyperplasia and basement membrane thickening,tubular epithelial cell swelling and deformation,shedding,mesangial cell proliferation. High-dose oat β-glucan could significantly increase the abundance and diversity of intestinal flora,the ratio of Firmicutes/Bacteroidetes in the diabetic nephropathy rats( P < 0. 05). Conclusion Oat β-glucan can improve renal function in diabetic rats,increase the abundance and diversity of intestinal flora and increase the ratio of Firmicutes/Bacteroidetes. This may be one of the mechanisms to delay diabetic nephropathy.
