响应面法优化超高压提取蓝莓花色苷工艺及其活性研究

【摘要】 为分析萃取特性和优化工艺参数,利用超高压提取技术从蓝莓中提取花色苷。采用单因素和Box-Behnken组合试验,考察提取压力、保压时间、乙醇体积分数和料液比对蓝莓花色苷含量的影响,并分析花色苷萃取特性;通过MTT法和Annexin V-FITC/PI双标记染色法分别研究蓝莓花色苷提取物对HepG2细胞增殖和凋亡的影响。结果表明超高压提取蓝莓花色苷最优工艺参数组合为:提取压力187MPa、保压时间6 min、乙醇体积分数57%、料液比1∶29(g/mL),在此条件下花色苷含量为(5.16±0.12)mg/g。体外抗肿瘤活性表明蓝莓花色苷提取物可通过抑制HepG2细胞增殖和促进其凋亡两种途径来达到抗肿瘤效果。该研究结果可为天然活性成分的提取提供一种有效方法,同时可为食品添加剂提供重要的资源。

【Abstract】 The ultra-high pressure extraction technology is used to extract anthocyanins from blueberry to analyze the extraction characteristics and optimize the process parameters.The effects of extraction pressure,pressure-holding time,ethanol volume fraction and solid-to-liquid ratio on the yield of anthocyanins from blueberry are investigated by single factor and Box-Behnken combination test,and the extraction characteristics of anthocyanins are also analyzed.The effects of blueberry anthocyanin extracts on the proliferation and apoptosis of HepG2 cells are studied by MTT assay and Annexin VFITC/PI double-labeled staining respectively.The results show that the optimal process parameters of ultra-high pressure extraction of anthocyanins from blueberry are as follows:the extraction pressure is 187 MPa,the pressure-holding time is 6 min,the ethanol volume fraction is 57%and the solid-to-liquid ratio is 1∶29(g/mL),under such conditions,the yield of anthocyanins is(5.16±0.12)mg/g.The anti-tumor activity in vitro indicates that the blueberry anthocyanin extracts could achieve antitumor effect by inhibiting the proliferation of HepG2 cells and promoting their apoptosis.The research results can provide an effective method for the extraction of natural active ingredients.At the same time,it can provide important resources for food additives.
