寺河矿15号煤层中产氢产乙酸微生物的富集和群落结构研究

【摘要】 煤炭地下生物气化是利用微生物在地下将煤中有机组分进行多级降解后将固态资源转化为气态资源的一种开采方法。前期研究表明,多数煤层产甲烷菌属于乙酸发酵型产甲烷菌,因此产乙酸菌在煤炭生物气化过程中发挥着承上启下的重要作用。以寺河矿15号煤层中微生物菌群作为研究对象,首先分别通过添加3种不同碳源的培养基对寺河矿煤层地质微生物系统中的产乙酸菌进行富集培育,并对其产酸产气特征进行系统分析;然后通过高通测序比较的分析富集过程中微生物群落结构,探究寺河矿煤层中可被富集培育的产乙酸菌类型;最后得出的研究结果表明,寺河矿煤层中可富集培育的产乙酸菌主要包括Clostridium SP.、Lachnoclostridiu SP.和Terrisporobacter SP.。最终的研究结论说明,寺河矿煤层中的产乙酸菌群由多种具备产乙酸特征但又各自具有独特代谢方式的菌属构成,并从乙酸生成途径进一步揭示出寺河矿煤层地质产甲烷菌群群落结构的多样性特征。

【Abstract】 Underground microbial coal gasification is a mining method that uses microorganisms to degrade organic components in coal and then convert solid resources into gaseous energy. Previous studies showed that most coal seam methanogens belonged to acetic acid fermentative methanogens, so acetogenic bacteria played an connecting role in the process of coal microbial gasification. The microbial community in Sihe No.15 coal seam is taken as the research object, three different carbon sources were added to culture medium to enrich and cultivate the acetogenic bacteria in Sihe coal geological microbial system, and the characteristics of acid and gas production were analyzed systematically; the microbial community structure in the enrichment process was analyzed by high-throughput sequencing, and the types of acetogenic bacteria that could be enriched and cultivated in Sihe coal seam were explored; the results showed that Clostridium SP., Lachnocltridiu SP. and Terrisporobacter SP. could be enriched and cultivated in Sihe coal seam. The final research conclusion showed that the acetogenic bacteria community in Sihe coal seam was composed of a variety of bacterial genus with acetic acid production characteristics but with unique metabolic mode, and the research further revealed the diversity characteristics of Sihe coal geological methanogenic bacteria community structure from the acetic acid production path.
