lncRNA SNHG16在人脑胶质瘤组织中的表达及其与病人预后的关系

【摘要】 目的探讨长链非编码RNA(lncRNA)SNHG16在人脑胶质瘤组织中的表达及其与病人预后的关系。方法实时荧光定量PCR检测104例2013年9月至2015年1月本院病理科冻存的胶质瘤组织和2018年4月至2019年1月颅脑损伤内减压术中切除的40例正常脑组织lncRNA SNHG16的相对表达量。以SNHG16表达量的中位数为截断值,将胶质瘤病人分为高表达组和低表达组。随访截止时间为2019年6月,记录总生存期(OS)和无进展生存期(PFS)。结果胶质瘤组织SNHG16相对表达量明显高于正常脑组织(P<0.001);高级别胶质瘤组织SNHG16相对表达量明显高于低级别胶质瘤组织(P<0.001)。多因素Cox比例回归风险模型分析结果显示,SNHG16高表达是胶质瘤病人OS和PFS缩短的独立危险因素(P<0.05)。生存曲线分析显示高表达组OS(中位数25个月,四分位数18~34个月)和PFS(中位数15个月,四分位数9~18个月)较低表达组(OS中位数35个月,四分位数20~42个月;PFS中位数19个月,四分位数12~20个月)明显缩短(P<0.05)。结论人脑胶质瘤SNHG16呈高表达,与病人不良生存预后和肿瘤进展有关。

【Abstract】 Objective To investigate the expression of long non-coding RNA(lncRNA) small nucleolar RNA host gene 16(SNHG16) in human glioma tissues and its relationship with the prognosis of glioma patients. Methods Real-time fluorescence quantitative PCR was used to detect the relative expression level of lncRNA SNHG16 in glioma tissues obtained from 104 patients with glioma who underwent mocirosurgery from September 2013 to January 2015 and in normal cerebral tissues obtained from 40 patients with traumatic brain injury who underwent decompression from April 2018 to January 2019. Taking the median of SNHG16 expression as the cut-off value, glioma patients were divided into high expression group and low expression group. The deadline for follow-up was June 2019, and overall survival(OS) and progression-free survival(PFS) were recorded. Results The relative expression level of SNHG16 in glioma tissues was significantly higher than that in normal cerebral tissues(P<0.05). The relative expression level of SNHG16 in high-grade glioma tissues was significantly higher than that in low-grade glioma tissues(P<0.05). The multivariate Cox proportional regression risk model analysis showed that high expression of SNHG16 was risk factors for shorter OS and PFS of glioma patients(P<0.05). The OS and PFS of the low expression group were significantly higher than those of the high expression group(P<0.05). Conclusions The lncRNA SNHG16 is highly expressed in human gliomas. The high expression of SNHG16 is related to the poor survival prognosis and tumor progression of glioma patients.
