鹌鹑性别的分子鉴定方法研究

【摘要】 为快速、准确鉴定出鹌鹑胚胎或组织样品的性别,研究以CHD1基因为候选基因,设计并合成了用于朝鲜鹌鹑性别鉴定的引物,建立了两种鉴定鹌鹑性别的分子生物学方法。第一种鉴定方法是对提取的鹌鹑基因组DNA进行PCR扩增,将扩增产物进行琼脂糖凝胶电泳,出现单一条带的个体为雄性鹌鹑;出现两条清晰条带的个体为雌性鹌鹑。第二种方法是利用荧光定量PCR进行扩增,接着进行熔解曲线分析,出现单个峰的为雄性鹌鹑;出现两个峰的为雌性鹌鹑。结果表明:这两种方法均能有效地进行鹌鹑性别的分子鉴定,其中第一种方法的结果直观可靠,可应用于鹌鹑的育种工作中;第二种方法操作简便,需要的时间短,适合实验室中大量组织样品(个体)性别的分子鉴定。

【Abstract】 In order to develop a quick and reliable method to identify gender of quail embryo or tissue sample, CHD1 gene was used as candidate gene to design primers for gender identification of Korean quail, and two molecular biological methods were established. The segments of CHD1 were amplified by PCR and analyzed by agarose gel electrophoresis in the first method. The sample shown single band was identified as coming from male quail, while sample shown double bands was identified as coming from female quail. The second method was to amplify CHD1 intron in fluorescence quantitative PCR, then the melting curve of PCR amplification products were analyzed. The sample shown single peak was identified as coming from male quail, and sample shown double peaks was identified as coming from female quail. The results showed that two methods could identify the gender of quail effectively, and the results of first method was clear and believable, which could be used in quail breeding; while the second method was easy and time-saving, which was suitable for the gender identification with large number of tissue samples or individuals in laboratory.
