MPTP致帕金森病小鼠嗅觉障碍的机制研究
The mechanism of olfactory impairment in MPTP-induced mouse model of Parkinson’s disease
【摘要】 目的观察1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPTP)致帕金森病(parkinson’s disease,PD)小鼠的嗅觉障碍并探讨其作用机制。方法采用MPTP小鼠模型,通过行为学、免疫组织化学和免疫印迹的方法观察MPTP小鼠的嗅觉障碍及其相关机制。结果行为学的结果显示MPTP小鼠的嗅觉能力是下降的,相关免疫组化和免疫印迹的结果显示MPTP小鼠嗅球中的多巴胺(dopamine,DA)能神经元的数量是上升的。结论 MPTP小鼠的嗅觉功能下降并且和嗅球中DA能神经元的上升相关。
【Abstract】 Objective To investigate the mechanism of olfactory impairment in 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine( MPTP)-treated mice. Methods Mice were treated with MPTP,the olfactory changes and their related mechanism in MPTP-treated mice were observed by behavioral,immunohistochemical and western blot methods. Results The results from praxiology experiment showed that the olfactory function of MPTP-treated mice was decreased,while the results from related immunohistochemistry and western blot showed that the number of dopaminergic neuron in the olfactory bulb of MPTP-treated mice was increased. Conclusion The olfactory dysfunction in MPTP-treated mice was associated with an increase in the number of olfactory bulb DA neurons.
