巴特日-7对肠炎大鼠及其血清TNF-α、IL-1β、IL-10水平的影响
Effects of Bateri-7 on the enteritis rats and their serum levels of TNF-α, IL-1β, and IL-10
【摘要】 目的探究巴特日-7对肠炎大鼠及其血清TNF-α、IL-1β、IL-10水平的影响。方法采用葡聚糖硫酸钠构建肠炎大鼠模型,90只SD大鼠随机分为正常对照组、模型组、阳性对照组、巴特日-7水提物组(低、中、高剂量),巴特日-7有效部位组(低、中、高剂量),肠炎大鼠造模第4天开始灌胃给药,每天1次,连续7 d。检测各组大鼠体质量、24 h的摄食量、疾病活动指数、结肠湿/干质量比,ELISA检测血清TNF-α、IL-1β、IL-10水平,HE染色观察结肠组织病理学改变。结果与模型组比较,巴特日-7水提物、巴特日-7有效部位大鼠体质量、24 h摄食量增加,疾病活动指数、结肠湿/干质量比降低,TNF-ɑ、IL-1β水平降低,IL-10水平增加(P<0.05,P<0.01),结肠组织病理学改善。结论巴特日-7能改善肠炎大鼠的症状,降低TNF-ɑ、IL-1β水平,升高IL-10水平。
【Abstract】 AIM To investigate the effects of Bateri-7 on the enteritis rats and their serum levels of TNF-α, IL-1β, and IL-10. METHODS Glucan sodium sulfate was used to induce a rat model of enteritis. Ninety SD rats were randomly divided into normal control group, model group, positive control group, Bateri-7 water extract groups(at low dose, medium dose and high dose, respectively), Bateri-7 differential extract groups(at low dose, medium dose and high dose, respectively). Enteritis rats were intragastrically administered with different agents once daily for 7 consecutive days from the 4 th day after the modelling. The rats were subjected to measurement of body mass, food intake, disease activity index(DAI) and colonic wet/dry weight ratio,the determination of serum TNF-α, IL-1βand IL-10 levels by ELISA, and observation of colonal histopathological changes by HE staining. RESULTS Compared with the model group, groups intervened with Bateri-7 water extract, or Bateri-7 differential extract had improved body bass and the food intake at 24 h, reduced index levels revealing disease activity and colon wet/dry weight ratio, decreased TNF-ɑ and IL-1β levels, increased IL-10 levels(P<0.05,P<0.01),and improved colon histopathology. CONCLUSION Bateri-7 can improve the symptoms of enteritis rats, reduce their levels of TNF-ɑ and IL-1β, and increase their IL-10 level.
