山慈菇含药血清对人乳腺癌 SK-BR-3细胞增殖、凋亡以及迁移能力影响的实验研究
Experimental Study on the Effect of Iphigenia indica Kunth Drug Serum on the Proliferation, Apoptosis and Migration Ability of Human Breast Cancer SK-BR-3 Cells
【摘要】 目的观察山慈菇含药血清对人乳腺SK-BR-3细胞增殖、凋亡以及迁移能力的影响。方法选用20只SD雌性大鼠,2月龄,体重(200±20)g,使用随机数字表法将大鼠随机分为实验组和空白对照组,每组10只,实验组大鼠按0.01 mL/g灌胃山慈菇浓煎剂,对照组大鼠给予等体积的生理盐水,每日灌胃1次,连续6d,末次灌胃后,腹主动脉取血,制作含药血清;同时培育人乳腺癌SK-BR-3细胞。使用MTT法检测人乳腺癌SK-BR-3细胞增殖情况、荧光显微镜观察SK-BR-3凋亡情况以及划痕试验检测SK-BR-3细胞迁移情况。结果与对照组比较,实验组能够明显抑制人乳腺癌SK-BR-3细胞的增殖、诱导细胞凋亡以及延缓人乳腺癌SK-BR-3细胞向划痕中央聚集。结论山慈菇含药血清能够有效抑制人乳腺癌SK-BR-3细胞活性,并加速细胞凋亡以及延缓细胞的迁移能力。
【Abstract】 Objective:To observe the effects of Iphigenia indica Kunth drug serum on the proliferation,apoptosis and migration ability of human breast SK-BR-3 cells. Methods:20 SD female rats of 2 months old and weighing(200 ± 20)g were randomly divided into an experimental group and a blank control group by using a random number table method,10 rats per group. The rats in the experimental group were administered with Iphigenia indica Kunth decoction and the rats in the control group were given the same volume of physiological saline once a day for 6 days. In the last time,the abdominal aorta was taken for blood to make drug serum. At the same time human breast cancer SK-BR-3 cells were bred. MTT method was used to detect the proliferation of the human breast cancer SK-BR-3 cells. SK-BR-3 apoptosis was observed by using a fluorescence microscope,and SK-BR-3 cell migration was detected by a scratch test. Results:Compared with the control group,the experimental group can significantly inhibit the proliferation of the human breast cancer SK-BR-3 cells,induce cell apoptosis and delay the accumulation of the human breast cancer SK-BR-3 cells toward the center of the scratch. Conclusion:Iphigenia indica Kunth drug serum can effectively inhibit human breast cancer SK-BR-3 cell activity,accelerate cell apoptosis and delay cell migration.
