间充质干细胞装载纳米化疗药向乳腺癌细胞递送

【摘要】 目的:本研究旨在验证间充质干细胞能装载一种纳米化疗药(Nano-DOX)并向乳腺癌细胞递送。方法:利用CCK-8及蛋白质免疫印迹法检测Nano-DOX对间充质干细胞活力的影响;利用激光共聚焦显微镜荧光分析及流式细胞仪检测Nano-DOX被间充质干细胞装载的情况;利用激光共聚焦显微镜荧光分析及流式细胞仪检测载药间充质干细胞向乳腺癌细胞的药物递送。结果:间充质干细胞可高效装载Nano-DOX并保持活力,乳腺癌细胞可促进载药间充质干细胞释放Nano-DOX并摄取之。结论:本研究表明间充质干细胞可装载纳米化疗药物Nano-DOX并向乳腺癌细胞递送。该策略为抗肿瘤药物的递送及利用间充质干细胞治疗肿瘤提供了新的思路。

【Abstract】 Objective This study aims to verify that mesenchymal stem cells(MSCs) can be loaded with a nano chemotherapy drug(Nano-DOX) and delivered to breast cancer cells. Methods CCK-8 and western blotting were used to detect the effect of Nano-DOX on the viability of mesenchymal stem cells. Laser confocal microscope fluorescence analysis and flow cytometry were used to detect the loading of Nano-DOX by MSCs. Confocal microscope fluorescence analysis and flow cytometry to detect drug delivery of drug-loaded MSCs to breast cancer cells. Results MSCs can efficiently load Nano-DOX and maintain vitality. Breast cancer cells can promote drug-loaded MSCs to release Nano-DOX and ingest it. Conclusion This study shows that mesenchymal stem cells can be loaded with Nano-DOX and delivered to breast cancer cells. This strategy provides a new idea for the delivery of anti-tumor drugs and the use of mesenchymal stem cells to treat tumors.
