双波长高效液相色谱法同时测定乌蕨中6种主要活性成分含量

【摘要】 目的建立同时测定乌蕨中原儿茶酸、原儿茶醛、荭草苷、牡荆素鼠李糖苷、牡荆素、木犀草苷等6种主要活性成分含量的双波长高效液相色谱法。方法选用赛默飞(Thermo)A型C18柱(4.6 mm×250 mm,5μm),以乙腈(A)-0.4%磷酸溶液(B)为流动相,梯度洗脱,原儿茶酸、原儿茶醛检测波长为280 nm,荭草苷、牡荆素鼠李糖苷、牡荆素、木犀草苷检测波长为350 nm。柱温:30℃,流速:1 mL·min-1;进样量:5μL。结果乌蕨中原儿茶酸、原儿茶醛、荭草苷、牡荆素鼠李糖苷、牡荆素、木犀草苷六种活性成分分离度良好,质量分别在0.161 9~1.214 4,0.063 7~0.477 9,0.083 5~0.626 5,0.238 5~1.788 5,0.077 8~0.583 7,0.026 3~0.197 0μg呈良好的线性关系,均r≥0.999 9,平均回收率分别为100.42%,101.78%,99.17%,105.12%,97.86%,101.54%;RSD分别为2.38%,2.53%,2.31%,3.06%,2.13%,3.87%。结论该方法简单,结果准确,重复性、稳定性好,可为制订乌蕨的质量控制标准提供参考依据。

【Abstract】 Objective A dual wavelength high performance liquid chromatography(HPLC) method was established for the simultaneous determination of protocatechuic acid, protocatechuic aldehyde, orientin, oramnosylvitexin, vitexin, and luteolin-7-o-glucoside in Stenoloma chusanum(L.) Ching. Methods The analysis was performed on a Thermo-Finnegan C18(250 mm×4.6 mm,5 μm) column with mobile phase of acetonitrile(A)-0.4% phosphoric acid solution(B) at the flow rate of 1.0 mL·min-1for gradient elution.The determintion wavelength was 280 nm for protocatechuic acid and protocatechui caldehyde, and 350 nm for orientin, oramnosylvitexin, vitexin and luteolin-7-o-glucoside.The column temperature was 30 ℃ and injection volume was 5 μL. Results The six compounds were well separated. The linear ranges were from 0.161 9 to 1.214 4, 0.0637 to 0.477 9, 0.083 5 to 0.626 5, 0.238 5 to 1.788 5, 0.077 8 to 0.583 7, and 0.0263 to 0.197 0 μg for protocatechuic acid, protocatechuic aldehydeorientin, orientin, oramnosylvitexin, vitexin, luteolin-7-o-glucoside, respectively,with the correlation of r≥0.999 9. The average recoveries were 100.42%(RSD 2.38%), 101.78%(RSD 2.53%), 99.17%(RSD 2.31%), 105.12%(RSD 3.06%), 97.86%(RSD 2.13%) and 101.54%(RSD 3.87%). Conclusion The method is simple, accurate, reproducible and stable, which can provide reference for the quality control standard of Stenoloma chusanum(L.)Ching.
