固相萃取结合高效液相色谱法测定甘草中6种有效成分的含量

【摘要】 目的建立固相萃取结合高效液相色谱法测定不同产地甘草中6种有效成分的定量方法。方法采用固相萃取法提取甘草中的有效成分,并用高效液相色谱法(HPLC)对其药效组分进行测定。流动相为乙腈-0.05%磷酸水溶液,梯度洗脱,流速为1 mL/min,检测波长为240 nm。结果甘草中6种药效组分芍药内酯苷、芍药苷、芹糖甘草苷、甘草苷、苯甲酰芍药苷、甘草酸分别在28.8~920.0、14.7~470.0、61.9~1980.0、48.4~1550.0、71.9~2300.0、27.5~880.0μg/mL范围内呈现良好的线性关系,平均回收率依次为98.44%、101.25%、96.78%、103.74%、97.85%、95.87%,RSD值分别为1.98%、3.02%、2.85%、1.99%、2.47%、2.06%。结论固相萃取结合HPLC法检测甘草有效成分快速、可靠。

【Abstract】 Objective To establish a quantitative method of solid phase extraction and high performance liquid chromatography in determination of six active ingredients in Glycyrrhizae Radix et Rhizoma from different habitats. Methods The effective constituent of Glycyrrhizae Radix et Rhizoma was extracted by solid phase extraction and its active ingredients were determined by high performance liquid chromatography(HPLC). The mobile phase was acetonitrile-0.05% phosphoric acid solution, with gradient elution, flow rate of 1 mL/min, and detection wavelength of 240 nm. Results The six active ingredients albiflorin, paeoniflorin, liquiritin apioside, liquiritin, benzoylpaeoniflorin, glycyrrhizic acid in Glycyrrhizae Radix et Rhizoma showed good linear relationship in the range of 28.8-920.0, 14.7-470.0, 61.9-1980.0, 48.4-1550.0, 71.9-2300.0, 27.5-880.0 μg/mL, the recovery rate was 98.44%, 101.25%, 96.78%, 103.74%,97.85%, 95.87% respectively, and the RSD was 1.98%, 3.02%, 2.85%, 1.99%, 2.47%, 2.06% respectively. Conclusion Solid phase extraction combined with HPLC in detection of the active ingredients of Glycyrrhizae Radix et Rhizoma is fast and reliable.
