【摘要】 目的检测Transgelin基因在A型主动脉夹层(TAAD)患者主动脉壁中的表达,分析其表达水平与主动脉夹层血管壁直径的相关性,预测Transgelin基因在主动脉夹层发病中的临床诊断价值。方法 2017～2018年于华中科技大学同济医学院附属同济医院采集13例TAAD患者的主动脉壁以及11例无夹层的主动脉壁(源自扩心病/冠心病患者心脏移植受体)作为对照,通过HE和EVG染色进行形态学检测,采用免疫组化、实时荧光定量PCR、以及蛋白质免疫印迹法检测Transgelin在对照和主动脉夹层血管壁中的表达和亚细胞定位情况。结果 Transgelin基因在TAAD患者的主动脉壁中表达显著升高,但其表达水平与升主动脉血管壁直径以及主动脉弓部直径无显著相关性。结论 Transgelin表达上调可能促进主动脉夹层的发生,有望成为预测主动脉夹层发生的生物学标志物。更多还原

【Abstract】 Objective To detect the expression of Transgelin gene in the aortic wall of patients with Type A aortic dissection(TAAD), analyze the correlation between the expression level of Transgelin and the diameter of aorta wall of TAAD patients, and predict the clinical diagnostic value of Transgelin in aortic dissection. Methods During the period of 2017 to 2018, thirteen cases of TAAD patients’ aorta tissues and eleven cases of control aorta tissues derived from heart transplantation patients with dilated cardiomyopathy or coronary heart disease were collected at Tongji Hospital affiliated to Tongji Medical College of Huazhong University of Science and Technology. Hematoxylin-eosin staining and Verhoeff′s Van Gieson staining were performed to detect morphological changes of aorta, and immunohistochemistry, real-time PCR, combined with Western blot were used to investigate the expression pattern of Transgelin gene and its subcellular localization in aorta tissues of TAAD and control individuals. Results The expression levels of Transgelin were significantly up-regulated in the aorta of TAAD patients, however, the expression levels of Transgelin were not correlated with the diameter of ascending aortic wall or aortic arch. Conclusion The up-regulation of Transgelin may promote the occurrence of aortic dissection. Moreover, Transgelin might be a potential biomarker for predicting the occurrence of aortic dissection.更多还原
