【摘要】 目的探讨成纤维细胞生长因子2(FGFR2)、人类表皮生长因子受体2(HER2)、张力蛋白同源磷酸酶基因(PTEN)在胃癌组织中的表达及临床意义。方法选取我院2015年1月～2018年12月行手术切除的胃癌组织标本84例,采用免疫组化方法检测胃癌组织中FGFR2、HER2及PTEN的表达,分析三个基因与胃癌临床病理特征的关系。结果 FGFR2、HER2在胃癌组织中的表达率为67.80%、46.40%,均高于正常胃黏膜组织,差异有统计学意义(P<0.05);PTEN在胃癌组织中的表达率为38.10%,低于正常胃黏膜组织,差异有统计学意义(P<0.05)。不同浸润深度、TNM分期、肿块大小的患者胃癌组织中FGFR2蛋白的表达比较,差异有统计学意义(P<0.05);不同浸润深度、肿块大小的患者胃癌组织中HER2的表达比较,差异有统计学意义(P<0.05);不同肿瘤分化程度、淋巴结转移的患者胃癌组织中PTEN的表达比较,差异有统计学意义(P<0.05);相关性分析显示,PTEN表达与FGFR2及HER2的表达均呈负相关(r=-0.233、-0.316),HER2与FGFR2的表达呈正相关(r=0.586)。结论FGFR2、HER2及PTEN与胃癌的发生发展密切相关,其中PTEN的表达与HER2、FGFR2负相关,HER2与FGFR2的表达呈正相关,联合检测三项基因表达对有助于胃癌的预后判断及靶向药物的选择。更多还原

【Abstract】 Objective To investigate the expression and clinical significance of fibroblast growth factor 2(FGFR2), human epidermal growth factor receptor 2(HER2) and tensin homophosphate gene(PTEN) in gastric cancer tissues.Methods From January2015 to December2018,84 cases of gastric cancer specimens which were surgically removedin our hospital were selected.The expression of FGFR2, HER2 and PTEN in gastric cancer was detected by immunohistochemistry, and its association withclinicopathological characteristics was analyzed. Results The expression rates of FGFR2 and HER2 in gastric cancer tissues were 67.80% and 46.40%, both higher than normal gastric mucosal tissues,the difference was statistically significant(P<0.05); the expression rate of PTEN in gastric cancer tissues was 38.10%, which was lower than In normal gastric mucosal tissue, the difference was statistically significant(P<0.05). Comparison of FGFR2 protein expression in gastric cancer tissues of patients with different invasion depths, TNM stages, and tumor sizes was statistically significant(P<0.05); comparison of HER2 expression in gastric cancer tissues of patients with different invasion depths and tumor sizes, the differences were statistically significant(P <0.05); Comparison of the expression of PTEN in gastric cancer tissues of patients with different tumor differentiation and lymph node metastasis, the difference was statistically significant( P<0.05);Correlation analysis showed that PTEN expression was negatively correlated with FGFR2 and HER2 expression(r =-0.233,-0.316), and HER2 and FGFR2 expression were positively correlated(r=0.586).Conclusion FGFR2、HER2 and PTEN are closely related to the occurrence and development of gastric cancer, in which the expression of PTEN is negatively correlated with HER2、FGFR2 and the expression of FGFR2 is positively correlated. Combined detection of three gene expression is helpful to the prognosis judgment of gastric cancer and the choice of targeted drugs.更多还原
