釉基质蛋白对异位骨再生影响的研究

【摘要】 目的探讨釉基质蛋白(EMPs)对正常大鼠和糖尿病大鼠异位骨再生的影响。方法采用改良乙酸法从幼猪的牙胚中提取EMPs并进行鉴定。建立正常大鼠和糖尿病大鼠双侧极量颅骨缺损模型,实验侧植入10mg/ml EMPs及其载体藻酸丙二醇酯(PGA),对照侧植入载体PGA。术后4周、8周行骨密度分析及组织学检测,探讨骨再生情况。结果骨密度分析显示正常大鼠在4周、8周实验侧骨密度均显著高于对照侧(P<0.05),糖尿病大鼠则在4周和8周时均无显著性差异。组织学分析显示正常大鼠在4周、8周时实验侧骨再生均优于对照侧;糖尿病大鼠在4周、8周时双侧均无骨再生发生。结论 10mg/ml EMPs可以促进正常大鼠颅骨缺损的骨再生,对糖尿病大鼠则无明显促进作用。EMPs对糖尿病大鼠骨再生的有效浓度和作用有待于进一步研究。

【Abstract】 Objective To investigate the effects of enamel matrix proteins(EMPs)on ectopic bone regeneration in normal and diabetic rats. Methods EMPs were extracted from the tooth germs of young pigs by modified acetic acid method and identified. A model of bilateral extreme skull defects in normal rats and diabetic rats was established. The experimental side was implanted with 10 mg/ml EMPs and its carrier propylene glycol alginate(PGA), and the control side was implanted with carrier PGA. Bone mineral density analysis and histological examination were performed 4 weeks and 8 weeks after surgery to explore bone regeneration. Results Bone mineral density analysis showed that the bone density of the experimental rats at 4 weeks and 8 weeks was significantly higher than that of the control side(P<0.05). However,there was no significant difference between the diabetic rats at 4 weeks and 8 weeks. Histological analysis showed that the normal rat bone regeneration was better than that of the control side at 4 and 8 weeks in normal rats; no bone regeneration occurred in both sides of diabetic rats at 4 and 8 weeks. Conclusion 10 mg/ml EMPs can promote bone regeneration in normal rat skull defects,but it has no obvious promoting effect on diabetic rats. The effective concentration and effect of EMPs on bone regeneration in diabetic rats need further study.
