藤梨根提取物对肝癌细胞生物学特性的影响以及机制研究

【摘要】 目的探讨藤梨根提取物对肝癌细胞生物学特性的影响以及潜在作用机制。方法通过不同浓度藤梨根正丁醇提取物(0,25,50,100,200,400μg/mL)作用于人肝癌HepG2细胞。采用噻唑蓝(MTT)法检测肝癌HepG2细胞增殖情况,采用流式细胞术和Transwell法检测细胞凋亡、迁移、侵袭情况,采用Western blot法检测核因子-κB(NF-κB)信号通路蛋白表达水平。结果不同浓度藤梨根提取物对人肝癌HepG2细胞增殖有抑制作用(P均<0.05),且具有浓度依赖性,其半数抑制浓度(IC50)为145.365μg/mL。145μg/mL的藤梨根提取物可显著抑制HepG2细胞迁移、侵袭(P<0.05),诱导凋亡(P<0.05),抑制NF-κB磷酸化(P<0.05),下调基质金属蛋白酶-2(MMP-2)蛋白水平(P<0.05)。结论藤梨根正丁醇提取物可能通过影响NF-κB信号通路调控肝癌细胞生物学特性。

【Abstract】 Objective It is to investigate the effects of kiwi root extract on the biological characteristics of hepatocellular carcinoma cells and its potential mechanism. Methods The human hepatocellular carcinoma HepG2 cells were treated with different concentrations of kiwi root extract(0, 25, 50, 100, 200, 400 μg/mL). The cell proliferation of HepG2 was detected by MTT. The cell apoptosis, invasion and migrationwere was detected by flow cytometry and Transwell assay. The expression level of nuclear factor-κB(NF-κB) signaling pathway protein were detected by Western blot. Results Different concentrations of kiwi root extract all could inhibited the proliferation of human hepatoma HepG2 cells(P<0.05) with a concentration-dependent manner. The half maximal inhibitory concertration(IC50) of kiwi root extract was 145.365 μg/mL. 145 μg/mL kiwi root extract could significantly inhibit the invasion and migration of HepG2 cells(P<0.05), and induced their apoptosis(P<0.05), it inhibited the phosphorylation of NF-κB and down-regulate the level of matrix metalloproteinase-2(MMP-2)(P<0.05). Conclusion The kiwi root extract could regulate the biological characteristics of hepatocellular carcinoma cells maybe by influencing NF-κB signal pathway.
