斑翅果蝇寄生蜂果蝇锤角细蜂大量饲养过程中寄主蛹分离方法的比较
Comparison of host pupae separation methods in mass-rearing of Trichopria drosophilae,a parasitoid of Drosophila suzukii
【摘要】 利用直接揉搓法、滑石粉法、玉米淀粉法、水洗法4种方法,分离斑翅果蝇(Drosophila suzukii Matsumura)的寄生蜂果蝇锤角细蜂(Trichopria drosophilae Perkins)在大量饲养过程中的替代寄主黑腹果蝇(Drosophila melanogaster Meigen)的蛹,并测定这些分离方法对黑腹果蝇蛹的自动计数、果蝇锤角细蜂搜寻寄主、黑腹果蝇的羽化、寄主蛹壳和蜂蛹的分离、果蝇锤角细蜂的羽化5个方面的影响。结果表明,黑腹果蝇蛹的黏连对自动数粒、风选分离、果蝇锤角细蜂的寄生率均有显著的负面影响;滑石粉法和玉米淀粉法对黑腹果蝇蛹的分离效果最佳,且对果蝇锤角细蜂的寄生率、出蜂率和寄生蜂与果蝇的总羽化率均无显著影响。
【Abstract】 Drosophila suzukii Matsumura is an important fruit pest. However,Trichopria drosophilae Perkins,a pupal parasitoid of D. suzukii has good potential for biological control of this pest. Drosophila melanogaster Meigen is used as an alternative host for the mass-rearing of T. drosophilae. In this study,methods including direct rubbing,talc,cornstarch,and water washing were used for D. melanogaster pupae separation. The efficacy of the above-mentioned methods was estimated using automatic pupae counting,host-searching of T. drosophilae,separation of host puparium and parasitoid pupae,and emergence of T. drosophilae. Our results showed that the agglomeration of D. melanogaster pupae had a significantly negative effect on the automatic pupae counts,winnowing separation,and parasitism rates of T. drosophilae. Based on our results,the talc and cornstarch methods were the best for D. melanogaster pupae separation. Neither method had a significant effect on parasitism rate,emergence rate of T. drosophilae,or total emergence rate of T. drosophilae and D. melanogaster.
