猪膀胱脱细胞基质制备的实验研究
Experimental Research of Method with Preparing Porcine Bladder Acelluar Matrix
【摘要】 目的研究猪膀胱脱细胞基质的制备方法。方法实验组使用200 m L 1mol/L Na Cl含DNAse-Ⅰ 2 000 k U和RNAse 2 000 k U的溶液,对照组使用200 m L 1 mol/L Na Cl含DNAse-Ⅰ 2 000 k U的溶液,将处理后的膀胱粘膜下基质,在37℃恒温振荡10 h,转速150次/min,PBS溶液反复清洗3次后放在100 m L 4%脱氧胆酸钠溶液中,37℃恒温振荡10 h,转速150次/min,进行2次,PBS溶液反复清洗3次。制备出2种猪膀胱脱细胞基质。实验组和对照组制备的基质进行病理切片,HE染色,显微镜下观察脱细胞基质效果。结果和结论实验组制备的猪膀胱脱细胞基质满足脱细胞技术要求。
【Abstract】 Objective This paper aims to study the preparation of porcine bladder acellular matrix.Methods 200 m L1 mol/L NaCl solution containing DNAse-Ⅰ 2 000 k U and RNase 2 000 k U was used in the experimental group,and 200 m L1 mol/L Na Cl solution containing DNAse-Ⅰ 2 000 k U was used in the control group. The treated bladder submucosal matrix was shaken at 37℃ for 10 h at a rotating speed of 150 times/min. PBS solution was repeatedly cleaned for 3 times,and then placed in 100 m L 4% sodium deoxycholate solution,and was shaken at 37℃ for 10 h. The rotating speed was 150 times/min,twice,and PBS solution was repeatedly cleaned for 3 times. Two kinds of acellular matrix of pig bladder were prepared. The experimental group and the control group prepared the matrix for pathological sections,HE staining,microscopic observation of the effect of acellular matrix. Results and conclusion The porcine bladder acellular matrix prepared in the experimental group met the requirements of acellular technology.
