泰山白首乌中白首乌二苯酮在大鼠体内药代动力学研究
Study on pharmacokinetic of Baishouwu benzophenone in rats after intragastric administration the extract of Cynanchum bungei Decne
【摘要】 目的建立大鼠血浆中白首乌二苯酮的HPLC测定方法,研究泰山白首乌中白首乌二苯酮在大鼠体内的药代动力学特征。方法 SD雄性大鼠灌胃给药后不同时间点眼内眦取血,血浆用乙酸乙酯处理后,用HPLC测定血浆中白首乌二苯酮的浓度,根据测定结果绘制血药浓度-时间曲线,利用DAS 2.0软件拟合药动学参数。结果白首乌二苯酮浓度在5.0~300.0μg/mL范围内,峰面积与浓度线性关系良好(R2=0.9966),平均回收率为(82.96±2.2)%,精密度、稳定性和专属性均符合有关规定。结论白首乌二苯酮在大鼠体内的药动学模型为二室模型,药动学参数Tmax=0.25 h,Cmax=249.46μg/mL,AUC(0-t)=980.47 mg/(L·h)。
【Abstract】 Objective: To establish A HPLC method for the determination of Baishouwu benzophenone in rat plasma to study the pharmacokinetic of Baishouwu benzophenone in rats after intragastric administration the extract of Cynanchum bungei Decne. Methods: Blood was collected from the inner canthus of SD male rats at different time points after intragastric administration, then the concentration of Baishouwu benzophenone in the ethyl acetate extract of plasma was determined with the proposed HPLC. the plasma concentration-time curve was made and the pharmacokinetic parameters were fitted using DAS 2.0 software. Results: There was a better liner relationship(R2=0.9966) between peak area and concentration(5.0~300 μg/mL). The average recovery was(82.96±2.2)%, precision and stability were in compliance with the regulations. Conclusion: The pharmacokinetic results showed Baishouwu benzophenone is fitted in a two-compartment model,pharmacokinetic parameters Tmax=0.25 h,Cmax= 249.46 μg/mL,AUC(0-t)= 980.47 mg/(L·h).
