牛樟芝滴丸对大鼠非酒精性脂肪肝相关基因表达的影响
Effects of Antrodia Camphorata Dropping Pill (牛樟芝滴丸) on Related Gene Expression of Rats with Non-Alcoholic Fatty Liver Disease
【摘要】 目的:研究牛樟芝滴丸对大鼠非酒精性脂肪肝(NAFLD)相关基因表达的影响。方法:SD大鼠48只,随机分成6组:正常对照组,模型组,水飞蓟素组(20 mg·kg-1),牛樟芝滴丸高、中、低剂量组(320、160、80 mg·kg-1),造模成功后给药6周。实验结束后,分别测定大鼠肝组织脂肪酸合成酶(FAS)、乙酰辅酶A羧化酶(ACC)、过氧化物酶体增殖物激活受体α(PPAR-α)、固醇调节元件结合蛋白1c(SREBP-1c)的m RNA和蛋白表达。结果:与模型组比较,牛樟芝滴丸低剂量组SREBP-1c mRNA和蛋白表达以及FAS蛋白表达显著降低(P <0. 05,P <0. 01),但FAS mRNA表达无显著差异; PPAR-α蛋白表达显著升高(P <0. 01),PPAR-αmRNA表达与ACC mRNA和蛋白表达均无统计学意义;中、高剂量组SREBP-1c、FAS mRNA和蛋白表达均显著降低(P <0. 05,P <0. 01),而PPAR-α、ACC mRNA和蛋白表达均显著升高(P <0. 01)。结论:牛樟芝滴丸对大鼠NAFLD的相关基因表达具有明显的影响。
【Abstract】 Objective: To investigate the effects of Antrodia Camphorata Dropping Pill on gene expression related to non-alcoholic fatty liver disease( NAFLD) in rats. Methods: A total of forty-eight SD rats were randomly divided into six groups: the normal control group,the model group,the silymarin group( 20 mg ·kg-1),the Antrodia Camphorata Dropping Pill groups of high,middle and low doses( 320,160,80 mg·kg-1).After modeling successfully,rats of each group were Ig the corresponding medicine for six weeks. The FAS,ACC,PPAR-α,SREBP-1 c m RNA and protein expressions were respectively determined. Results: Compared with the model group,the SREBP-1 c m RNA and protein expressions and the FAS protein expression in the Antrodia Camphorata Dropping Pill groups of low doses were significantly decreased( P < 0. 05,P < 0. 01),but the FAS m RNA expression was not significant difference,the PPAR-α protein expression was significantly increased( P < 0. 01),the PPAR-α m RNA expression and the ACC m RNA and protein expression were not statistically significant; the expressions of SREBP-1 c and FAS m RNA and protein in the middle and high dose groups were significantly decreased( P < 0. 05,P < 0. 01),while the expressions of PPAR-α and ACC m RNA and protein were significantly increased( P < 0. 01). Conclusion: There was a significant effects of the Antrodia Camphorata Dropping Pill on gene expressions related to on NAFLD in rats.
