水貂FGF5基因多态性及其与毛长性状的关联分析
The Polymorphism of FGF5 Gene in Mink and Its Relationship Analysis with Hair Length
【摘要】 本研究旨在检测水貂成纤维细胞生长因子5(fibroblast growth factor 5, FGF5)基因多态性,解析该基因变异位点与水貂毛长性状的相关性。首先对30只美国短毛黑水貂的FGF5基因部分序列进行了扩增和测序;使用DNAMAN软件分析发现,FGF5基因内含子1中T376C位点存在多态性位点;通过在线酶切软件NEBcutter V2.0分析表明,Taq I内切酶可用于该SNP位点基因分型。以美国短毛黑水貂和丹麦咖啡水貂共计252个血液样本的基因组DNA作为模板,采用PCR-RFLP技术对252只美国短毛黑水貂和丹麦咖啡水貂FGF5基因T376C位点进行了基因分型,并将该位点与水貂毛长性状进行关联分析。结果表明:PCR扩增片段在376位点存在突变,该位点存在2个等位基因T和C,形成TT、TC和CC 3种基因型,该变异位点在美国短毛黑水貂群体中为中度多态(0.25 <PIC <0.05),而在丹麦咖啡水貂群体中表现为高度多态(PIC> 0.25);X~2检验结果显示,T376C位点在美国短毛黑水貂群体处于Hardy-Weinberg平衡状态,在丹麦咖啡水貂群体显著偏离Hardy-Weinberg平衡状态;关联分析表明,FGF5基因T376C位点与水貂毛长性状极显著相关(P <0.000 1)。研究结果提示,FGF5基因T376C位点可能是影响水貂毛长性状的主控位点,可作为水貂毛长性状潜在的分子遗传标记。
【Abstract】 This paper is to detect the polymorphism of FGF5 gene in mink and analyze the relationship between its variation sites and the hair length of mink.Firstly, the partial sequences of FGF5 gene of 30 American short-haired black minks were amplified and sequenced.The analysis results by DNAMAN software revealed that there was a polymorphic site at the T376 C site in intron 1 of FGF5 gene.Using NEBcutter V2.0 online enzymatic design software, it was found that Taq I could identify SNP.The blood genomic DNA of 252 samples from American short-haired black mink and Denmark coffee mink were used as templates, the genotype of T376 C mutation sites of FGF5 gene in 252 samples were identified by using PCR-RFLP technology.And the relationship between this site and the hair length of mink was analyzed.The results showed that PCR amplified fragments had mutation of 376 sites, there were two alleles T and C on this site, and three genotypes of TT, TC and CC were formed.T376 C locus polymorphism was at moderate polymorphic status in American short-haired black mink(0.25 < PIC < 0.05), but it was at high polymorphic status in Denmark coffee mink(PIC > 0.25).The results of Chi-square test showed that T376 C locus in American short-haired black mink was in Hardy-Weinberg equilibrium,which deviated significantly in Denmark coffee mink.The relationship analysis revealed that the T376 C locus polymorphism of FGF5 gene had extremely significant correlation with hair length of mink.The research results suggested that T376 C locus might affect hair length phenotype of mink, it could be used as potential molecular genetic marker of hair length phenotype of mink.
