组织培养中不同的消毒处理对白及种子萌发的影响

【摘要】 目的为了解决白及自然繁殖率低,提高白及种子的萌发率。方法该研究以成熟白及蒴果为实验材料,运用植物组织培养技术,采用单因素实验法和Box-Behnken响应面法,探究不同的消毒剂、消毒时间和摇床振荡频率等因素对白及种子萌发率的影响。结果消毒剂种类是影响白及种子消毒效果的重要因素;通过响应面法对消毒组合进行优化,可得最优的消毒组合是0.1%升汞,消毒时间10min,摇床振荡频率140Hz,利用该消毒组合,白及种子萌发率可达79.77%。结论优化筛选出来的消毒组合,可为白及的组织培养和工业化生产提供科学参考。

【Abstract】 Objective In order to solve the low reproduction rate of Bletilla striata, and improve the seed germination rate of Bletilla striata. Methods In this study, capsule fruits of Bletilla striata were used as experimental materials. Using plant tissue culture technology, single factor experiment and Box-Behnken response surface method were used to explore the effect of the factors such as different disinfectant, disinfection time, and frequency of shaker oscillation on the seed germination of Bletilla striata. Results The type of disinfectant was an important factor affecting the disinfection effect of Bletilla striata seed. Box-Behnken response surface method was used to optimize the disinfection combination treatment. The optimal disinfection combination was 0.1% HgCl2, disinfection time was 10 min, and the shaking frequency was 140 Hz. With the disinfection combination treatment, the seed germination rate of Bletilla striata could reach 79.77%. Conclusion The optimized disinfection combination can provide scientific reference for tissue culture and industrial production of Bletilla striata.
