福美加对去势骨质疏松大鼠长骨干骺端骨组织计量学和生物力学的影响

【摘要】 目的探讨福美加(阿仑膦酸钠维D3)对去势骨质疏松大鼠长骨干骺端骨组织计量学和生物力学的影响。方法 30只3月龄雌性SD大鼠随机分为假手术组(Sham)、去卵巢组(OVX)和福美加组(ALN)。福美加治疗12周后,对所有大鼠进行骨密度测定,收集大鼠股骨和胫骨,用于骨组织计量学和生物力学指标的测定。结果 ALN组股骨和L5椎体骨密度、胫骨骨质量参数(BV/TV、Tb.N、Tb.Th)明显大于OVX组(P <0.05)。Sham和ALN组股骨干及远端干骺端的最大载荷、破坏载荷和刚度明显高于OVX组(P <0.05)。OVX组与ALN组股骨干骺端弯曲试验最大载荷、破坏载荷的均值差异显著高于股骨干弯曲试验(P <0.05)。结论福美加可显著提高去势骨质疏松大鼠的骨密度、骨组织计量学参数和生物力学性能,且作用于长骨干骺端的效果大于长骨骨干。

【Abstract】 ObjectiveTo investigate the effect of alendronate vitamin D3 on histomorphometry and biome-chanics of long bone epiphysis in ovariectomized rats with osteoporosis.Methods Thirty 3-month-old female Sprague-Dawley rats were divided into group Sham,OVX and ALN. The rats in group OVX and ALN were bilaterally ovariectomized. After 12 weeks of treatment with alendronate vitamin D3,bone mineral density was measured in all rats,and femur and tibia were collected for the histomorphometry and biomechanics measurements.Results The BMD of femur and L5 vertebrae and bone mass parameters(BV/TV,Tb.N,Tb.Th)in group ALN was significantly higher than group OVX(P < 0.05). The maximum load,failure load,and stiffness at femoral diaphysis and distal femoral metaphysis in group Sham and ALN were significantly higher than that in group OVX(P < 0.05). The mean differences of maximum load and failure load between OVX and ALN group in metaphysis bending test were significantly higher than that in diaphysis bending test(P < 0.05).Conclusion Alendronate vitamin D3 could significantly improve the BMD,histomorphometry and biomechanics in ovariectomized rats with osteoporosis,and the effect on the metaphysis is greater than diaphysis.
